龙葵成分茄解啶对人大肠癌SW620细胞凋亡和AKT-GSK3B信号的转导作用 被引量:3

摘要 目的:研究龙葵成分茄解啶对人大肠癌SW260细胞的作用及机制。方法:茄解啶作用SW620细胞,CCK-8法检测细胞增殖,Annexin V-FITC/PI双染、流式细胞仪检测细胞凋亡,试剂盒检测Caspase-3、Caspase-8、Caspase-9活性,Western blot检测蛋白表达。结果:茄解啶可抑制SW620细胞增殖,诱导细胞凋亡,活化Caspase-3、Caspase-8、Caspase-9,抑制AKT表达及其磷酸化,上调GSK3B表达,抑制GSK3B磷酸化,降低CCND1、c-Myc表达,升高Bad表达,抑制Bad磷酸化。结论:茄解啶可抑制SW620细胞增殖并诱导其凋亡,其机制与调控AKT-GSK3B信号转导相关。 Objective:To study the effects and mechanism of purapuridine(PPD),a compound in Solanum nigrum,on human colorectal cancer SW260 cells.Methods:SW620 cells were treated with PPD.Cell proliferation was detected by CCK-8 assay,apoptosis was detected by flow cytometry with Annexin V-FITC/PI double staining,and protein expression was detected by Western blot.Results:PPD inhibited SW620 cells proliferation,induced cell apoptosis,activated Caspase-3,Caspase-8 and Caspase-9,down-regulated AKT expression and phosphorylation,increased GSK3 B expression,inhibited GSK3 B phosphorylation,decreased CCND1 and c-Myc expressions,increased Bad expression,and inhibited Bad phosphorylation.Conclusion:PPD inhibits cell proliferation and induces apoptosis in SW620 cells,the mechanism may be related to AKT-GSK3 B signal transduction.
