摘要 目的:对壮药毛果鱼藤Derris eriocarpa How的化学成分进行分离鉴定,并对分离到的化合物进行抗肿瘤活性评价。方法:采用硅胶、Sephadex LH-20葡聚糖凝胶、ODS柱色谱等方法对毛果鱼藤的化学成分进行分离纯化,运用理化性质和现代波谱学技术进行化合物的结构鉴定;采用MTT法对化合物进行抗肿瘤活性评价。结果:从毛果鱼藤乙酸乙酯萃取物中分离得到12个化合物,分别鉴定为:3,5-二甲氧基-4′-异丙基反式二苯乙烯(1)、2′-deoxyisoauriculatin(2)、flemichapparin B(3)、1-[6-羟基-2-甲氧基-2″,2″-二甲基吡喃-(5″,6″∶3,4)]-2-(4′-甲氧基苯基)-1,2-乙二酮(4)、1-[6-羟基-2-甲氧基-2″,2″-二甲基吡喃-(5″,6″∶3,4)]-2-(3′,4′-亚甲二氧基苯基)-1,2-乙二酮(5)、dimethylalpinum isoflavone(6)、4-羟基-3-[4′-O-(3,3-二甲基烯丙基)苯基]-5-甲氧基-2″,2″-二甲基吡喃-(5″,6″∶6,7)-香豆素(7)、derrubone(8)、甘草宁G(9)、derrusnin(10)、木豆素(11)、6a-dihydroxy-9-methoxypterocarpan(12)。体外抗肿瘤活性筛选的结果显示,化合物8抑制Hela细胞的IC_(50)为10.62μmol/L,化合物9抑制HepG_(2)细胞的IC_(50)为56.90μmol/L。结论:其中,化合物2~6和9为首次从毛果鱼藤中分离得到,化合物1、11和12为首次从鱼藤属植物中分离得到。化合物8具有较好的肿瘤细胞生长抑制作用,化合物9具有较弱的抗肿瘤活性。 Objective:To isolate and identify the chemical constituents of the Zhuang Medicine Derris eriocarpa rattan,and to evaluate the antitumor activity of the isolated compounds.Methods:The compounds were isolated and purified by silica gel,Sephadex LH-20,ODS column chromatography and others,the structures were identified according to the physicochemical properties and modern spectroscopy;The antitumor activities of the compounds were evaluated by MTT method.Results:Twelve compounds were isolated from the ethyl acetate fraction of Derris eriocarpa rattan and identified as 3,5-dimethoxy-4′-hidroxy-3′-prenyl-trans-stilbene(1),2′-deoxyisoauriculatin(2),flemichapparin B(3),1-[6-hydroxy-2-methoxy-2″,2″-dimethylpyrano-(5″,6″∶3,4)]-2-(4′-methoxyphenyl)-1,2-ethanedione(4),1-[6-hydroxy-2-methoxy-2″,2″-dimethylpyrano-(5″,6″∶3,4)]-2-(3′,4′-methylenedioxyphenyl)-1,2-ethanedione(5),dimethylalpinum isoflavone(6),4-hydroxy-3-[4′-O-(3,3-dimethylallyl)phenyl]-5-methoxy-2″,2″-dimethylpyrano-(5″,6″∶6,7)-coumarin(7),derrubone(8),gancaonin G(9),derrusnin(10),cajanin(11),6 a-dihydroxy-9-methoxypterocarpan(12),respectively.The results of in vitro anti-tumor activity screening showed that compound 8 inhibited the growth of Hela cells with IC_(50)values of 10.62μmol/L,and compound 9 inhibited the growth of HepG_(2)cancer cells with IC_(50)values of 56.90μmol/L.Conclusion:Among them,compounds 2 to 6,9 are isolated from Derris eriocarpa for the first time.Compounds 1,11 and 12 are isolated from Derris genus for the first time.Compound 8 shows good tumor cell growth inhibition and compound 9 shows weak antitumor activity.
