摘要
目的:研究连翘Forsythia suspensa(Thunb.)Vahl的化学成分。方法:利用大孔吸附树脂、Sephadex LH-20葡聚糖凝胶、半制备高效液相色谱等分离技术进行分离纯化,通过理化性质及波谱数据解析结构。结果:从连翘的乙醇提取物中分离得到13个化合物,分别鉴定为:(R)-2-(3,4-二羟基苯基)-2-乙氧基乙酸甲酯(1)、4-羟基苯乙酸(2)、酪醇(3)、松柏苷(4)、甲基松柏苷(5)、3,4′-dihydroxy-propiophenone-3-β-D-glucopyranoside(6)、β-hydroxypropiovanillone-3-O-β-D-glucopyranoside(7)、N-(1′-D-去氧木糖醇基)-6,7-二甲基-1,4-二氢-2,3-喹喔啉二酮(8)、forsythenside H(9)、萹蓄苷(10)、山柰酚-3-O-α-L-鼠李糖-(1→6)-β-D-半乳糖苷(11)、(-)-开环异落叶松脂素-4-O-β-D-葡萄糖苷(12)、rel-5-(3S,8S-dihydroxy-1R,5S-dimethyl-7-oxa-6-oxobicyclo[3,2,1]-oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid(13)。结论:化合物1为新化合物,通过比较比旋光度的方法确定了其绝对构型。
Objective:To study the chemical constituents from the fruits of Forsythia suspensa.Methods:The various chromatographic techniques such as macroporous resin,Sephadex LH-20,semi-preparative HPLC column chromatography were used for isolated and purified,the structures were identified by physical and chemical properties and spectral data.Results:Thirteen compounds were isolated from the ethanol extract of the fruits of Forsythia suspensa and identified as methyl(R)-2-(3,4-dihydroxyphenyl)-2-ethoxyacetate(1),4-hydroxyphenylacetic acid(2),tyrosol(3),coniferin(4),methylconiferin(5),3,4′-dihydroxy-propiophenone-3-β-D-glucopyranoside(6),β-hydroxypropiovanillone-3-O-β-D-glucopyranoside(7),N-(1′-D-deoxyxylitolyl)-6,7-dimethyl-1,4-dihydro-2,3-quinoxalinedione(8),forsythenside H(9),avicularin(10),kaempferol-3-O-α-L-rhamnopyranosyl(1→6)-β-D-galactopyranoside(11),(-)-secoisolariciresinol-4-O-β-D-glucoside(12),rel-5-(3S,8S-dihydroxy-1R,5S-dimethyl-7-oxa-6-oxobicyclo[3,2,1]-oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid(13),respectively.Conclusion:Compound 1 is a new compound and its absolute configuration is confirmed by comparing the specific rotation.
