罂粟壳与蜜罂粟壳水煎液中5个生物碱类成分的对比研究 被引量:6

摘要 目的:建立HPLC法同时测定罂粟壳和蜜罂粟壳中5个生物碱类成分(可待因、吗啡、罂粟碱、蒂巴因和那可丁)的含量,并对含量进行比较。方法:采用资生堂CAPCELL PAK C_(18)(250 mm×4.6 mm,5μm)色谱柱;流动相为乙腈-含0.1%甲酸的0.02 mol/L乙酸铵溶液,梯度洗脱;检测波长为250 nm;流速为1.0 mL/min;柱温为30℃;进样量为10μL。结果:建立的5个生物碱类成分的含量测定方法简便、可行,罂粟壳与蜜罂粟壳中的可待因和罂粟碱含量差异显著,水煎后2种饮片中的5个生物碱类成分含量均有显著差异,且水煎后5个生物碱类成分的含量明显降低,蒂巴因的转移率最高,分别为46.6%、61.4%,那可丁的转移率最低,分别为12.9%、13.7%。结论:罂粟壳在蜜制后5个生物碱类成分含量升高,但水煎后含量降低。药典中以吗啡的绝对含量制定限量有待进一步完善。 Objective:To establish a HPLC method for the determination of the contents of five alkaloids(codein,morphine,papaverine,thebaine and narcotine)in Papaveris Pericarpium and honey-stir-baked Papaveris Pericarpium,and to compare the contents.Methods:The CAPCELL PAK C_(18)(250 mm×4.6 mm,5μm)column was used.The mobile phase was acetonitrile-0.02 mol/L ammonium acetate solution containing 0.1%formic acid with gradient elution,detection wavelength was 250 nm,flow rate was 1.0 mL/min,column temperature was 30℃,injection volume was 10μL.Results:Established five alkaloids component content determination method was simple,feasible.There were significant differences in the contents of codeine and papaverine in the Papaveris Pericarpium and honey-stir-baked Papaveris Pericarpium.The contents of five alkaloids in the two decoction pieces were significantly different after decocting,and significantly reduced after decocting.The transfer rate of thebaine was the highest,about 46.6%,61.4%,respectively,and the transfer rate of narcotine was the lowest,about 12.9%,13.7%,respectively.Conclusion:The contents of five alkaloids in Papaveris Pericarpium are increased after honey preparation,but decreased after decocting.Further improvement is needed to establish absolute limits for morphine in pharmacopoeia.
