摘要
目的:利用GC-MS代谢组学技术研究不同生长年限地骨皮初级代谢产物的差异,并进行代谢通路的富集,探讨初级代谢产物的差异代谢机制。方法:采用柱前衍生化GC-MS法测定样品,并结合聚类热图、偏最小二乘判别分析(PLS-DA)法对不同生长年限地骨皮的初级代谢产物进行多元统计分析。结果:根据VIP>1.0,P<0.05的原则,筛选得到3个高度相关的差异标志物,分别为天冬酰胺、柠檬酸和L-脯氨酸;通过MetaboAnalyst、KEGG等工具及代谢数据库进行代谢路径分析显示,精氨酸和脯氨酸代谢、丙氨酸、天冬氨酸和谷氨酸代谢、柠檬酸循环是不同生长年限地骨皮的主要差异代谢通路。结论:该研究探讨了不同生长年限地骨皮的初级代谢产物,为确定地骨皮最佳采收期提供了依据。
Objective:To study the differences of primary metabolites in Lycii Cortex with different growth years based on GC-MS metabolomics technique,to enrich the metabolic pathways and explore the differences in the metabolic mechanism of primary metabolites.Methods:The samples were determined by GC-MS method with pre-column derivation,and multivariate statistical analysis was performed for the primary metabolites of Lycii Cortex with different growth years by cluster heat map and partial least squares discriminant analysis(PLS-DA).Results:According to the principle of VIP>1.0,P<0.05,three highly correlated differential markers were screened,which were asparagine,citric acid and L-proline.Metabolic pathway analysis using Metaboanalyst,KEGG and other tools and metabolic database showed that the arginine and proline metabolism,alanine,aspartic acid and glutamate metabolism and citric acid cycle were the main metabolic pathways of Lycii Cortex with different growth years.Conclusion:In this study,the primary metabolites of different growth years of Lycii Cortex are studied,which can provide scientific basis for determining the best harvest time of Lycii Cortex.
