基于代谢组学分析酸枣仁贮藏期间化学成分的变化情况 被引量:1

Study on the Chemical Composition Changes of Ziziphi Spinosae Semen during Storage Periods Based on Metabonomics

摘要 目的:分析酸枣仁常规阴冷库贮藏1年前后的初级代谢产物和次级代谢产物的变化情况。方法:分别采用^(1)H-NMR法分析酸枣仁中初级代谢产物、UHPLC-Q-Orbitrap-MS法分析次级代谢产物,以主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)等多元统计方法为手段,找到贮藏前后酸枣仁样品中差异性的初级代谢产物和次级代谢产物。结果:^(1)H-NMR图谱确定了30个初级代谢产物;OPLS-DA分析显示贮藏1年后酸枣仁中7个初级代谢产物为差异成分。贮藏1年后半胱氨酸、甜菜碱和蔗糖的相对峰面积显著降低,而赖氨酸、乙酸、天冬酰胺和棉籽糖的相对峰面积显著升高。LC-MS结合多元统计分析共鉴定了17个差异次级代谢产物。结论:酸枣仁常规阴冷库贮藏1年后初级代谢产物和次级代谢产物相对含量均发生了显著的变化,该研究可为酸枣仁的贮藏有效期研究提供实验依据。 Objective:To analyze the changes of primary and secondary metabolites of Ziziphi Spinosae Semen stored in conventional cold storage for one year.Methods:Primary and secondary metabolites were analyzed by^(1)H-NMR method and UHPLC-Q-Orbitrap-MS method,respectively.Principal component analysis(PCA),orthogonal partial least squares discriminant analysis(OPLS-DA)and other multivariate statistical methods were used to find the differences of primary and secondary metabolites in Ziziphi Spinosae Semen before and after storage.Results:Thirty primary metabolites were annotated from^(1)H-NMR,and OPLS-DA analysis showed that seven primary metabolites in Ziziphi Spinosae Semen after storage for one year were different components.After storage for one year,the relative peak areas of cysteine,betaine and sucrose decreased significantly,while the relative peak areas of lysine,acetic acid,asparagine and raffinose increased significantly.A total of seventeen different secondary metabolites were identified by LC-MS combined with multivariate statistical analysis.Conclusion:The relative contents of primary and secondary metabolites of Ziziphi Spinosae Semen change significantly after storage in conventional cold storage for one year.This study can provide experimental basis for studying the storage validity of Ziziphi Spinosae Semen.
