基于网络药理学探索健脾解毒方治疗肝癌的机制 被引量:3

Study on the Mechanism of Jianpi Jiedu Prescription in Treating Hepatic Carcinoma Based on Network Pharmacology

摘要 目的:基于网络药理学方法分析健脾解毒方治疗肝癌的分子机制。方法:运用系统药理学方法查找并筛选健脾解毒方中药的活性成分及靶点,查找肝癌相关基因,确定健脾解毒方治疗肝癌的靶点,并进行富集分析以预测其可能的信号通路,探究其分子机制。结果:筛选得到健脾解毒方148种活性成分及与肝癌相关的靶点210个,依据拓扑学属性筛选出78个关键靶点,GO和Pathway富集分析发现其功能主要与细胞凋亡、细胞衰老、DNA转录、对缺氧的反应等有关,富集在PI3K-Akt通路、p53信号通路、IL-17信号通路、TNF信号通路、细胞凋亡通路等。结论:健脾解毒方治疗肝癌具有多途径、多靶点、多层次的特征,主要通过调节细胞增殖和凋亡及干预其肿瘤微环境等发挥抗肝癌作用,PI3K-Akt通路可能是其核心通路之一。 Objective:To analyze the molecular mechanism of Jianpi jiedu prescription in the treatment of hepatic carcinoma based on network pharmacology.Methods:The active components and targets of traditional Chinese medicine in Jianpi jiedu prescription were searched and screened by systematic pharmacology method,and the hepatocellular carcinoma related genes were searched to determine the targets of Jianpi jiedu prescription in treating hepatocellular carcinoma,and enrichment analysis was conducted to predict its possible signaling pathways and explore its molecular mechanism.Results:After screening,148 active components of Jianpi jiedu prescription and 210 targets related to hepatic carcinoma were obtained.According to the topological properties,78 key targets were selected.GO and Pathway enrichment analysis showed that its functions were mainly related to cell apoptosis,cell senescence,DNA transcription and response to hypoxia,and it was enriched in the PI3 K-Akt pathway,p53 signaling pathway,IL-17 signaling pathway,TNF signaling pathway,apoptosis pathway,etc.Conclusion:Jianpi jiedu prescription has characteristics of multi-pathways,multi-targets and multi-levels in treating hepatic carcinoma.It mainly plays an anti-liver cancer role by regulating cell proliferation and apoptosis and interfering with the tumor.The PI3 K-Akt pathway may be one of its core pathways.
