化学成分结合外观性状的赤芍质量等级标准研究

摘要 目的:建立同时测定赤芍中成分含量的方法,并分析化学成分与色度值、外观性状(根长度、直径)的相关性,探讨传统规格等级划分方法的合理性。方法:采用HPLC法测定儿茶素、氧化芍药苷、芍药苷、没食子酸、芍药内酯苷5个成分的含量;游标卡尺测量根长度、根直径;色差仪测定断面、表皮的颜色,以其为指标考察赤芍的外观性状特征;通过SPSS 21.0软件分析化学成分含量与外观性状的相关性,并进行聚类分析,建立赤芍的质量等级划分标准。结果:没食子酸的含量与色度值L^(*)、a^(*)、b^(*)呈极显著负相关(P<0.01),儿茶素、氧化芍药苷、芍药苷的含量与之呈极显著正相关(P<0.01),而各成分含量与根直径、根长度无显著相关性;21批赤芍样品可聚为2类。结论:该方法可同时测定赤芍中成分的含量;颜色越亮越红的赤芍样品中儿茶素、氧化芍药苷、芍药苷含量越高,没食子酸含量越低,而根直径、根长度与各成分含量无相关性。 Objective:To establish a method for simultaneous concent determination of components in Paeoniae Radix Rubra,and to analyze the correlation between chemical components and chromatic values,appearance characters(root length and root diameter),and to explore the rationality of the traditional standard grading method.Methods:The contents of catechin,oxypaeoniflorin,paeoniflorin,gallic acid and albiflorin were determined by HPLC.Vernier caliper was used to measure the length and diameter of roots;The colors of cross section and epidermal were measured by chromaticity difference analyzer,and the appearance characters of Paeoniae Radix Rubra were studied by using them as the indexes.SPSS 21.0 software was used to analyze the correlation between chemical component contents and appearance characters,and cluster analysis was conducted to establish the quality grade classification standard of Paeoniae Radix Rubra.Results:The content of gallic acid was negatively correlated with L^(*),a^(*),b^(*)(P<0.01),the contents of catechin,oxypaeoniflorin and paeoniflorin were significantly positively correlated with L^(*),a^(*),b^(*)(P<0.01),but there was no correlation between the component contents and diameter,length of roots;Twenty-one batches of samples of Paeoniae Radix Rubra could be clustered into two groups.Conclusion:This method can simultaneously determine the contents of the components in Paeoniae Radix Rubra.The brighter and redder the color,the higher the contents of catechin,oxypaeoniflorin and paeoniflorin in Paeoniae Radix Rubra,the lower the content of gallic acid,while the diameter and length of roots had no correlation with the content of each component.
