摘要
目的:基于网络药理学方法探讨青风藤治疗类风湿关节炎(RA)的可能机制。方法:通过TCMSP平台筛选青风藤活性成分,利用UniProt数据库将相应靶点蛋白转换为标准蛋白基因名,经Cytoscape 3.7.1软件构建网络图。在GeneCards数据库、OMIM数据库、CTD数据库中检索“Rheumatoid Arthritis”,获取RA疾病靶点,利用韦恩图得到药物成分和RA共同靶点,进行PPI网络分析,GO富集和KEGG通路富集分析。结果:获取青风藤6种成分及58个潜在靶点,RA疾病2060个相关靶点,青风藤-RA共同靶点25个,PPI网络中自由度靠前的3个靶点基因为CASP3、JUN、PTGS2,GO富集结果为408个和KEGG通路富集为20条信号通路。结论:青风藤可能是通过调节CASP3、JUN、PTGS2等靶点,发挥抑制炎症反应、调节免疫功能及调控细胞凋亡等作用来治疗RA。
Objective:To investigate the possible mechanism of Sinomenii Caulis in the treatment of rheumatoid arthritis(RA)by network pharmacology.Methods:The active components of Sinomenii Caulis were screened by TCMSP platform.The corresponding target proteins were converted into standard protein gene names using UniProt database,and Cytoscape 3.7.1 software was used to construct the network diagram.Rheumatoid Arthritis was searched in GeneCards database,OMIM database and CTD database,the disease targets of RA were obtained,the common targets of drug components and RA were obtained by using the Wayne diagram.PPI network analysis,GO enrichment analysis and KEGG pathway enrichment analysis were performed.Results:A total of 6 components and 58 potential targets of Sinomenii Caulis were obtained,2060 related targets of RA disease and 25 common targets between Sinomenii Caulis and RA were obtained.In the PPI network,the top 3 of target genes were CASP3,JUN and PTGS2.The GO enrichment results were 408 and KEGG pathways were enriched into 20 signaling pathways.Conclusion:Sinomenii Caulis may treat RA by regulating targets such as CASP3,JUN and PTGS2,inhibiting inflammatory response,regulating immune function and regulating cell apoptosis.
