大秦艽汤UPLC指纹图谱及8种成分含量测定研究 被引量:1

摘要 目的:建立大秦艽汤UPLC指纹图谱,并同时测定其中8个成分的含量。方法:采用ACQUITY UPLC^(■)BEH C_(18)(100 mm×2.1 mm,1.7μm)色谱柱;以乙腈-0.1%磷酸溶液为流动相,梯度洗脱;流速为0.4 mL/min;检测波长为240 nm;柱温为33℃;进样量为2μL。结果:建立了大秦艽汤UPLC指纹图谱,确定39个共有峰,并指认了11个成分,15批样品相似度均≥0.969。马钱苷酸、龙胆苦苷、升麻素苷、芍药苷、阿魏酸、甘草苷、黄芩苷、甘草酸8个成分在相应浓度范围内与峰面积的线性关系良好,r>0.9990。平均加样回收率为93.98%~99.18%,RSD为1.27%~3.81%。结论:建立的大秦艽汤UPLC指纹图谱及含量测定方法简便,稳定性、重复性良好,可为大秦艽汤的质量控制提供参考。 Objective:To establish the UPLC fingerprint of Daqinjiao decoction and to determine the contents of 8 components simultaneously.Methods:Using ACQUITY UPLC^(■)BEH C_(18)(100 mm×2.1 mm,1.7μm)column,the mobile phase was acetonitrile-0.1%phosphoric acid solution with gradient elution,flow rate was 0.4 mL/min,detection wavelength was 240 nm,column temperature was 33℃,injection volume was 2μL.Results:The UPLC fingerprint of Daqinjiao decoction was established,39 common peaks were identified and 11 components were identified.The similarity of 15 batches of samples was 0.969 or greater.The linear relationship between the 8 components,including loganic acid,gentiopicroside,cimicifugoside,paeoniflorin,ferulic acid,liquiritin,baicalin,glycyrrhizic acid,peak areas was good within the corresponding concentration range,r>0.9990.The average recovery rate was 93.98%~99.18%and RSD was 1.27%~3.81%.Conclusion:The established UPLC fingerprint and content determination method of Daqinjiao decoction is simple,stable and reproducibility,which can provide reference for quality control of Daqinjiao decoction.
