七味广枣散HPLC指纹图谱研究 被引量:3

摘要 目的:建立七味广枣散HPLC指纹图谱,并进行化学模式识别研究。方法:色谱柱为Alltech-Alltima C18(250 mm×4.6 mm,5μm)柱;流动相为甲醇-0.4%乙酸溶液,梯度洗脱;流速为0.8 mL/min;检测波长为254 nm;柱温为30℃;进样量为10μL;采用"中药色谱指纹图谱相似度评价系统(2012版)"进行相似度评价,采用SPSS 19.0、SIMCA-P 14.1软件进行聚类分析、主成分分析和正交偏最小二乘判别分析。结果:建立的指纹图谱共确定27个共有峰,指认了没食子酸、原儿茶酸、5-羟甲基糠醛、鞣花酸、芦丁、丁香酚6个成分。聚类分析将样品聚为3类,结合主成分分析、正交偏最小二乘判别分析发现其中15个成分是造成不同批次样品间差异性的主要标志物。结论:该研究建立的HPLC指纹图谱可为七味广枣散(丸)的质量评价及进一步研究提供参考。 Objective:To establish the HPLC fingerprint of Qiwei guangzao powder,and to carry out the chemical pattern recognition study.Methods:The study was carried out on an Alltech-Alltima C18(250 mm×4.6 mm,5μm)column with mobile phase consisted of methanol-0.4%acetic acid solution(gradient elution)at the flow rate of 0.8 mL/min,the detection wavelength was 254 nm,column temperature was 30℃,injection volume was 10μL.The similarity of 10 batches of samples was evaluated by"Similarity evaluation system of chromatographic fingerprint of TCM(2012 edition)".Cluster analysis,principal component analysis and orthogonal partial least squares discriminant analysis were performed by using SPSS 19.0 and SIMCA-P 14.1.Results:The established fingerprints had a total of 27 common peaks and 6 components of them were identified as gallic acid,protocatechuic acid,5-hydroxymethylfurfural,ellagic acid,rutin and eugenol.Cluster analysis grouped the samples into 3 categories,combined with principal component analysis and orthogonal partial least squares discriminant analysis,15 of them were found to be the main markers that caused the differences of different batches of samples.Conclusion:The HPLC fingerprint established in this study can provide reference for the quality evaluation and further research of Qiwei guangzao powder or Qiwei guangzao pill.
