摘要
目的:通过收集ACE/AngⅡ/AT1R通路基因,运用网络药理学方法,挖掘具潜在价值的有效中药和活性化合物。方法:通过KEGG、TCMSP数据库检索靶点基因、相关的有效中药以及活性化合物,运用Cytoscape 3.7.0构建有效中药-活性化合物-靶点网络,通过Network Analyzer功能分析各组分网络维度参数。结果:共有185味中药含该通路靶点基因,排名前10位为柴胡、桂枝、防风、辣椒、前胡、牛膝、土茯苓、余甘子、小茴香、紫花前胡。进一步构建有效中药-活性化合物-靶点网络,发现以土茯苓、柴胡、余甘子的Degree最高(Degree=3),活性化合物按Degree排序以oleic acid(Degree=8)、acetaldehyde(Degree=8)、acetic acid(Degree=5)居前3。结论:本研究初步分析了针对ACE/AngⅡ/AT1R通路上的有效中药,并通过网络药理学分析,为进一步探索治疗COVID-19,筛选了具潜在价值的中药及化合物。
Objective:To excavate the effective traditional Chinese medicine and active compounds with potential value by collecting ACE/AngⅡ/AT1 R pathway genes and using network pharmacology method.Methods:KEGG and TCMSP databases were used to retrieve target genes,relevant effective traditional Chinese medicines and active compounds.Cytoscape 3.7.0 was used to construct an effective traditional Chinese medicines-active compounds-targets network,the network dimension parameters of each component were analyzed by Network Analyzer.Results:A total of 185 traditional Chinese medicines contained the target genes of this pathway,and the top 10 were as follows,Bupleuri Radix,Cinnamomi Ramulus,Saposhnikoviae Radix,Capsici Fructus,Peucedani Radix,Achyranthis Bidentatae Radix,Smilacis Glabrae Rhixoma,Phyllanthi Fructus,Foeniculi Fructus and Peucedanidecursivi Radix.The effective traditional Chinese medicines-active compounds-targets network was constructed to detect that the Degree of Smilacis Glabrae Rhixoma,Bupleuri Radix and Phyllanthi Fructus was the highest(Degree=3)in traditional Chinese medicines,while oleic acid(Degree=8),acetaldehyde(Degree=8)and acetic acid(Degree=5)were the top 3 active ingredients in degree.Conclusion:This study preliminary analyses the effective traditional Chinese medicine for the ACE/AngⅡ/AT1 R pathway,and a network pharmacologic analysis is conducted to screen potentially valuable traditional Chinese mediciness and ingredients for further exploration of the treatment of COVID-19.
