基于网络药理学的辛夷-苍耳子药对治疗鼻咽癌作用机制探析 被引量:6

The Treatment Mechanism of Magnoliae Flos and Xanthii Fructus Herb Pairs on Nasopharyngeal Carcinoma based on Network Pharmacology

摘要 目的:探究辛夷-苍耳子药对治疗鼻咽癌(NPC)的有效活性化合物及其靶点的作用机制。方法:本研究通过TCMSP数据库筛选出辛夷-苍耳子药对中的化合物及相关靶点蛋白,并运用Uniprot数据库和Genecard数据库查询与靶点蛋白相对应的人类基因名称,进行标准化处理;通过Genecards数据库挖掘鼻咽癌疾病靶点,将其与中药靶点蛋白映射;将映射出的药对治疗NPC的有效靶点导入Cytoscape 3.2.1构建网络图,并运用CTD数据库进行GO富集和KEGG通路富集。结果:获得辛夷-苍耳子药对治疗NPC的共22个成分和49个有效靶点;获得17条GO生物过程和13条相关信号通路,涉及MAPK通路(MAPK signaling pathway)、PI3K-Akt信号通路(PI3K-Akt signaling pathway)等。结论:辛夷-苍耳子药对活性靶点主要通过MAPK信号通路、PI3K-Akt信号通路、MicroRNAs信号通路调控鼻咽癌细胞过程及其代谢来发挥治疗作用。 Objective:To explore the effect mechanism of Magnoliae Flos and Xanthii Fructus herb pairs on the effective active compounds and their targets in the treatment of nasopharyngeal carcinoma(NPC).Methods:The active components and targets of Magnoliae Flos and Xanthii Fructus herb pairs were screened through TCMSP database.Uniprot database and Genecard database were used to query the names of human genes corresponding to the target protein and to conduct standardization.Genecards database was used to mine nasopharyngeal cancer disease targets and map them with TCM target proteins.The mapped drug was introduced into Cytoscape 3.2.1 to construct the network map of effective targets for NPC treatment,and CTD database was used for GO enrichment and KEGG pathway enrichment.Results:A total of 22 components and 49 effective targets of Magnoliae Flos and Xanthii Fructus herb pairs for treating NPC were obtained,17 GO biological processes and 13 related signaling pathways were obtained,including MAPK signaling pathway and PI3 K-Akt signaling pathway.Conclusion:Magnoliae Flos and Xanthii Fructus herb pairs plays a therapeutic role on active targets mainly through MAPK signaling pathway,PI3 K-Akt signaling pathway and MicroRNAs signaling pathway in regulating the process and metabolism of nasopharyngeal carcinoma cells.
