华重楼2种叶斑病病原菌分离与鉴定 被引量:6

摘要 目的:鉴定引起四川地区华重楼焦斑病和圆纹病的病原菌。方法:采用组织分离法对病原菌进行分离,利用科赫氏法则验证其致病性,依据菌株的形态学和基于rDNA-ITS、TEF和Tub2基因序列分析对病原菌进行鉴定。结果:华重楼焦斑病病原菌的ITS-Tub2基因序列与已登录的Leptosphaerulina australis(登录号分别为MH861604、GU237540)聚类在一个分支上,支持率为100%;圆纹病病原菌的ITS-TEF基因序列与已登录的Clonostachys rosea(登录号分别为KC806254、KX184997)的同源性为100%。结论:根据病原菌的形态学和分子生物学,将引起华重楼焦斑病病原菌鉴定为小米壳属真菌Leptosphaerulina australis,圆纹病病原菌鉴定为链孢粘帚霉属真菌Clonostachys rosea。 Objective:To identify the pathogen causing burnt spot and ring spot of Paris polyphylla var.chinensis in Sichuan province.Methods:The pathogens were isolated by tissue isolation method,pathogenicity was verified by Koch′s rule.The pathogen was identified based on morphological characteristics and rDNA-ITS,TEF and Tub2 gene sequence analysis.Results:The ITS-Tub2 genes sequences of pathogens causing burnt spot in Paris polyphylla var.chinensis was 100% homology with Leptosphaerulina australis(No.MH861604,GU237540),clustering on a branch.The ITS-TEF gene sequences of pathogens causing ring spot in Paris polyphylla var.chinensis showed 100% homology with Clonostachys rosea(No.KC806254,KX184997).Conclusion:Based on the morphology and molecular biology of pathogen,Leptosphaerulina australis is identified as the pathogen causing burnt spot and Clonostachys rosea is identified as the pathogen causing the ring spot in Paris polyphylla var.chinensis.
