摘要
目的:采用网络药理学方法探讨四物汤的功效物质基础和药理机制。方法:依托中药系统药理学分析平台(TCMSP)检索四物汤4味中药的化学成分和作用靶点。采用Cytoscape软件构建化合物-靶点网络和靶点-通路网络,依据在线STRING数据库构建蛋白质相互作用(PPI)网络,研究四物汤药理作用的机制。结果:通过筛选获得24个化合物,相应靶点84个。PPI网络包含69个靶点,关键靶点涉及JUN、IL6、PIK3CG、TNF、PTGS2等。基因本体(GO)条目37个,其中生物过程相关的条目7个,分子功能相关的条目14个,细胞组成相关的条目13个。京都基因与基因组百科全书(KEGG)通路5条,涉及神经活性配体-受体、Ca2+、TNF、胆碱能突触、脂肪细胞脂解等信号通路。结论:本研究结果初步验证了四物汤的基本药理作用及其机制,为进一步深入探究其药理作用奠定了基础。
Objective:To explore the material basis and pharmacological mechanism of Siwu decoction by network pharmacology method.Methods:Based on TCMSP,the chemical constituents and target of Siwu decoction were retrieved.Cytoscape software was used to construct compound-target network and target-pathway network.Protein-protein interaction(PPI)network was constructed based on online STRING database to study the pharmacological mechanism of Siwu decoction.Results:Twenty-four compounds were obtained by screening with 84 corresponding targets.The PPI network contained 69 targets.The key targets involved JUN,IL6,PIK3 CG,TNF,PTGS2 and so on.There were 37 items of gene ontology(GO),including 7 items related to biological processes,14 items related to molecular functions and 13 items related to cell composition.Kyoto encyclopedia of genes and genomes(KEGG)contained five pathways,involving neuroactive ligand-receptor,Ca2+,TNF,cholinergic synapse,fat cell lipolysis signaling pathways.Conclusion:The results preliminarily verify the basic pharmacological effect and mechanism of Siwu decoction,lay a foundation for further study of its pharmacological effect.
