基于色差原理分析黄柏有效成分含量与颜色的相关性 被引量:24

摘要 目的:通过测定黄柏色差值及有效成分黄柏碱和小檗碱的含量,将代表颜色的外观指标值与代表质量的内在含量指标值相关联,探讨有效成分含量与颜色之间的相关性,为常用中药黄柏的质量评价提供参考依据。方法:利用色差仪对黄柏进行测定,以2015年版中国药典黄柏含量测定项下方法,采用HPLC进行测定,通过SPSS 21.0软件进行统计分析。结果:黄柏中黄柏碱、小檗碱与颜色指标值a~*(代表颜色红绿色度)、L~*(代表颜色亮度)、b~*(代表颜色黄蓝色度)、E~*ab(色差值)存在显著或极显著正相关关系(P<0.05或P<0.01),表明在一定程度上,颜色指标值L~*、a~*、b~*、E~*ab值越大,黄柏中黄柏碱、小檗碱含量越高。结论:黄柏药材颜色与有效成分小檗碱以及黄柏碱含量密切相关,且颜色偏亮黄色的黄柏有效成分含量较高,质量相对较好,通过色差仪对黄柏色差值进行测定可快速预测黄柏中有效成分的含量。 Objective:To study the correlation between the index value of appearance representing color and the index value of intrinsic content representing quality by measuring the color values of Phellodendron chinense and the contents of phellodendrine and berberine,and to explore the correlation between the contents of active ingredients and the color values,so as to provide basis for the quality evaluation of Phellodendron chinense.Methods:The color difference values of Phellodendron chinense was measured by colorimeter.The content determination method of phellodendrine and berberine in the 2015 edition of China Pharmacopoeia was used by HPLC,and statistical analysis was carried out by SPSS 21.0 software.Results:Phellodendrine and berberine contents in Phellodendron chinense were positively or extremely positively correlated with color index value a~*(representing color red-green direction),L~*(representing colorshade),b~*(representing color yellow-blue direction)and E~*ab(representing total color difference)(P<0.05 or P<0.01),indicating that to some extent,the higher the color index value L~*,a~*,b~*and E~*ab,the higher the contents of phellodendrine and berberine in Phellodendron chinense.Conclusion:The color of Phellodendron chinense is closely related to the contents of berine and phellodendrine,and the content of active ingredients in Phellodendron chinense with bright yellow color is higher and the quality is better,the determination of chromatic aberration value of Phellodendron chinense can quickly predicte the content of active ingredients in Phellodendron chinense.
