摘要
目的探讨RNA恒温核酸扩增法在儿童重症肺炎病原体检测中临床意义。方法选取我院收治的重症肺炎疾病患儿60例,分别采用PCR荧光探针法和RNA恒温核酸扩增法(SAT)检测患儿肺炎病原体,对比两组检测方式的检测检出率。所有患儿采用相同方式治疗,分析RNA恒温核酸扩增法同儿童重症肺炎严重程度的相关性。结果RNA恒温核酸扩增法检测出病原体患儿59例,检出率为98.33%,PCR荧光探针法检测出病原体患儿58例,检出率为96.67%,两者差异无统计学意义(P>0.05);在RNA恒温核酸扩增法转阴时间逐渐延长后,患儿发热持续时间显著延长,淋巴细胞百分比降低,中性粒细胞百分比升高,差异有统计学意义(P<0.05)。RNA恒温核酸扩增法灵敏度为100.00%,特异度为50.00%。结论在检测儿童重症肺炎病原体时,PCR荧光探针法和RNA恒温核酸扩增法,检出率相当,RNA恒温核酸扩增法对于肺炎发展以及预后可以进行有效评估,灵敏度较高,可以作为一种新型检测儿童重症肺炎病原体方式。
Objective To explore the clinical significance of RNA thermostatic nucleic acid amplification in pathogen detection of children with severe pneumonia.Methods A total of 60 children with severe pneumonia disease in our hospital were selected.PCR fluorescent probe method and RNA thermostatic nucleic acid amplification method were used to detect pneumonia pathogens in children,and the detection rates in two groups through the two detection methods were compared.All children were treated with the same methods,and the correlation between RNA thermostatic nucleic acid amplification and the severity of severe pneumonia in children was analyzed.Results Fiftynine cases of pathogens were detected by RNA thermostatic nucleic acid amplification,the detection rate was 98.33%;and 58 cases of pathogens were detected by PCR fluorescent probe,the detection rate was(96.67%);the difference was not statistically significant(P>0.05).The duration of fever was significantly prolonged,the percentage of lymphocytes was decreased,and the percentage of neutrophils was increased,showing a significant difference(P<0.05)after the time of turning negative by the method of RNA thermostatic nucleic acid amplification was prolonged;the difference was not statistically significant(P>0.05).The sensitivity and specificity of the RNA thermostatic nucleic acid amplification method were found to be 100.00%and 50.00%,respectively.Conclusion Upon detection of severe pneumonia in children pathogens,the use of PCR fluorescence probe method is used for testing and RNA detection detection rate isothermal nucleic acid amplification method,RNA isothermal nucleic acid amplification method to the development of pneumonia and healing can effectively evaluate,to obtain high sensitivity,can be used as a new type of detection of severe pneumonia in children pathogens.
