摘要
以50份抗大豆花叶病毒3号株系的大豆品种为实验材料,利用36对SSR引物,对供试材料进行遗传多样性分析及指纹图谱的建立。结果表明:36对引物具有稳定的多态性,供试材料共检测出191对等位基因,平均等位变异数为5.31,多态信息含量变化范围为0.2984~0.8072。采用UPGMA方法进行聚类分析,50份抗源品种间遗传相似系数在0.6387~0.8901之间,在遗传相似系数0.715处,分为3个大类群;在遗传相似系数0.74处,又将第三大类群分为6个亚群。根据每对引物多态信息含量的高低,进一步筛选出4对核心引物,构建了50份抗源品种的指纹图谱。遗传相似系数较小、聚在不同类群的抗源大豆品种,可能含有不同的抗性基因。
50 varieties resistant to soybean mosaic virus strain 3 were used as experimental materials,36 pairs of SSR primers were used to analyze the genetic diversity of the tested materials and construct fingerprints.The results showed that 36 pairs of primers had stable polymorphism,and a total of 191 alleles were detected in the tested materials.The average allelic variation was 5.31,and the polymorphic information content ranged from 0.2984 to 0.8072.The UPGMA method was used for cluster analysis.The genetic similarity coefficient among 50 resistant varieties was between 0.6387 and 0.8901.The 50 test materials were divided into 3 large groups at the genetic similarity coefficient of 0.715.At the genetic similarity coefficient of 0.74,the third largest group was divided into six subgroups.According to the level of polymorphism information of each pair of primers,4 pairs of core primers were further screened,and 50 fingerprints of resistant varieties were constructed.The genetic similarity coefficient is small,and the anti-sources clustered in different groups of soybean may contain different resistance genes.
