基于OCT影像的激光致小鼠皮肤损伤的无创综合评估的初步研究

摘要 目的在体无创光学影像通过量化相关参数实现激光致皮肤损伤效果的综合评估。方法使用光学相干断层成像(OCT)系统对皮肤损伤进行无创、无辐射成像,并计算光学衰减系数评估皮肤损伤,将损伤区域三维可视化并分割量化,最后将病理图像与OCT影像进行对比分析创面的形态结构变化。结果本研究实现了OCT影像的光学衰减系数量化评估不同剂量超连续谱激光和中红外3.8μm激光的皮肤损伤效果,量化的光学衰减系数图谱反映出激光皮肤损伤区域的位置及面积信息,通过三维OCT影像将不同剂量激光致小鼠皮肤损伤的量效关系直观呈现;在不同剂量激光皮肤损伤后,其病理图像与OCT影像均显示出皮肤损伤斑内部层状结构不明显。结论本文提出了一种基于光学影像的激光皮肤损伤的无创综合评估方法,通过量化的OCT影像参数实现了激光皮肤损伤的评估,且通过三维影像直观实时呈现损伤;该量化分析方法在激光生物学效应探究及激光消融治疗中具有重要的应用潜力。 Objective In vivo noninvasive optical imaging can be used to evaluate the laser-induced skin damage by quantifying related parameters.Methods Optical coherence tomography(OCT)was used to perform noninvasively and radiation-free image on mouse skin burns burned by laser radiation.The optical attenuation coefficient(OAC)was computed through a Fourier domain method.The three dimensional(3-D)visualization of OCT images was implemented through a ray-tracing method to displaying burned.Furthermore,pathological sections were compared with OCT images to analyze the morphological change of the wound.Results This method comprehensively evaluated the skin burns from different doses of super continuum laser and mid-infrared 3.8μm laser.The quantified OAC mapping reflected the location and area information of the laser skin damage.The 3D visualization of OCT images presented the burned lesions induced by different doses of the lasers.Both OCT images and pathological sections showed that the layered structure inside the lesion was not obvious for different laser doses.Conclusions This study presents a noninvasive comprehensive evaluation method for laser-induced skin damage based on optical image.The quantified OCT image parameters realize the evaluation of the laser skin burns which are displayed in real time through 3D visualization.The proposed method,which uses optical imaging to quantify laser-induced skin burn lesions,has application potential in the exploration of laser-biological effect and laser ablation.
