摘要
建立HPLC法测定中药甘草中甘草查尔酮A含量的方法。首先确定HPLC法测定甘草查尔酮A的流动相、波长和理论板数,后通过单因素筛选和正交试验确定供试品制备方法。结果表明,HPLC法测定甘草中甘草查尔酮A的流动相为甲醇-水(70∶30),波长379 nm,理论板数按甘草查尔酮A峰计算应不低于6000。供试品的制备方法为取本品粉末约0.25 g,精密称定,加入甲醇100 m L,密塞,超声处理(功率250 W,频率99 kHz)60 min,冷却,用甲醇补足减失的质量,摇匀,滤过,即得。结果表明,本研究所建立的方法操作简便、结果准确、重复性好。
This study established a method for content of licochalcone A of Radix Glycyrrhizae by HPLC.First,the mobile phase composition,wavelength,theoretical plate were determined by HPLC,and then sample preparation were determined by single factor test and orthogonal test.The result showed that the mobile phase composition was methanol and wate(70∶30),wavelength was 379 nm,the number of theoretical plates calculated for licochalcone A peak was not less than 6000.The preparation of sample was to take powder about 0.25 g,weighed accurately,placed it in a corked conical flask,and then added100 mL methanol,pluged,weighed accurately,ultrasound(250 W,99 kHz)for 60 min,cooling,maked up the lost weight with methanol,shaked well,filtrated.As a result,this method was easy to operate and had eminent repeatability.
