高温胁迫下香菇基因组甲基化差异分析 被引量:4

摘要 高温是香菇生长发育过程中经常遇到的逆境因子。通过以香菇耐热菌株和热敏感菌株为研究对象,利用MSAP技术分析了两者在高温胁迫后基因组DNA甲基化水平和甲基化模式变化。结果表明,2个香菇菌株在热处理后基因组甲基化水平都表现为上升,但耐热菌株甲基化水平变化大于热敏感菌株;2个菌株DNA同时发生了甲基化和去甲基化现象,但变化模式存在差异。耐热菌株主要发生甲基化,而热敏感菌株主要发生去甲基化。 High temperature stress can influence the growth of Lentinula edodes mycelium,which affect the yield of L.edodes.In this study,the heat-resistant strain and heat-sensitive strain of L.edodes were used as materials to analyze the methylation level and methylation patterns of genomic DNA after high temperature stress by MSAP technique.The results showed that the methylation level of both shiitake strains increased after heat treatment,but the methylation level of heatresistant strain changed more than that of heat-sensitive strain.DNA methylation and demethylation happened in both strains at the same time,but the changing patterns were different,the heat-resistant strain mainly occurred methylation,while heat-sensitive strain mainly occurred demethylation.
