新城疫病毒利用网格蛋白和小窝蛋白受体途径感染鸡骨髓来源树突状细胞

Infection of Chicken Bone Marrow-Derived Dendritic Cells with Newcastle Disease Virus through Clathrin and Caveolin Receptor Pathways

摘要 新城疫病毒(NDV)可以通过多种内吞途径感染细胞,且其入胞途径可能具有细胞类型特异性。本研究以鸡骨髓源树突状细胞(chBM-DCs)为模型,探究NDV入侵该细胞的内吞途径。首先利用CPZ(CME抑制剂)和Dynasore(dynamin1/2抑制剂)预处理DCs,然后感染NDV强毒株Herts/33,通过Western blot和IFA检测NDV NP的胞内表达水平变化。药物预处理组NP的表达水平均显著降低,表明NDV能够通过网格蛋白介导的内吞途径进入DCs。进一步利用甲基-β-环糊精(MβCD)预处理DCs,然后感染Herts/33,通过Western blot和IFA检测NDV NP的胞内表达水平变化。结果显示药物预处理组NP的表达水平显著降低,表明NDV能够通过小窝蛋白介导的内吞途径进入DCs。本研究为全面阐明NDV感染宿主细胞的途径提供了理论基础。 Newcastle disease virus(NDV)can infect cells through various endocytosis pathways and its entry may be cell type specific.In this study,chicken bone marrow-derived dendritic cells(chBM-DCs)were used as models to explore the endocytosis pathways of NDV entering DCs.Firstly,DCs were pre-treated with CPZ(CME inhibitor)and Dynasore(dynamin1/2 inhibitor),respectively and then infected with the NDV virulent strain Herts/33.The intracellular expression levels of NDV NP were examined by Western blot and IFA.The expression levels of NP in the drug pretreatment groups were significantly reduced,indicating that NDV could enter DCs through clathrin-mediated endocytosis.The DCs were further pre-treated with methyl-β-cyclodextrin(MβCD)and then infected with Herts/33.The intracellular expression levels of NDV NP were detected by Western blot and IFA.The results showed that the expression levels of NP in the drug pretreatment groups were significantly reduced,indicating that NDV could enter DCs through caveolin-mediated endocytosis.This study provided experimental data for clarifying the pathways in which NDV infected host cells.
