摘要 转录因子BTB-CNC同源体1(BACH1)在大多数哺乳动物组织中广泛表达,在氧化应激、细胞周期、血红素稳态、炎症和免疫等方面起到关键的调节作用。近年来关于BACH1在心血管疾病、干细胞多能性维持和肿瘤等方面作用的研究有了突破性的进展。全基因组关联研究提示BACH1与心血管疾病密切相关。BACH1参与缺血性疾病后血管新生、高血压、动脉粥样硬化等多种心血管疾病的发生和发展。BACH1是维持干细胞干性和中内胚层分化过程中的关键因子。BACH1通过重编程肿瘤代谢以及改变上皮间充质转化表型,促进肿瘤增殖转移,同时可能通过铁死亡抑制肿瘤生长,在肿瘤中有双重功能。BACH1作为一个转录因子,有调控自身表达的能力,并且对下游靶基因具有转录激活和转录抑制作用。细胞表型和状态的不同、体内环境以及共调节因子的招募均会对BACH1的转录产生影响。文章对BACH1研究进展进行综述。 The transcription factor BTB and CNC homology 1(BACH1)is expressed widely in most mammalian tissues,functioning as a key regulator in oxidative stress-response,cell cycle,heme homeostasis,inflammation,and immunity.In recent years,great progress has been made in the research of BACH1 in cardiovascular diseases,stem cell pluripotency maintenance and cancer.Genome-wide association studies(GWAS)suggest a closely connection between BACH1 and cardiovascular diseases.BACH1 is involved in the occurrence and development of various cardiovascular diseases,such as ischemic angiogenesis,hypertension and atherosclerosis.BACH1 is a critical factor of stem cell identity maintenance and mesendodermal differentiation.BACH1 promotes tumor proliferation and metastasis by reprogramming tumor metabolism and altering epithelial-mesenchymal transformation phenotypes.While BACH1 may inhibit tumor growth by controlling ferroptosis,thus playing a dual role in tumors.BACH1 has the ability to regulate its own expression,and function as both a transcriptional activator and a transcriptional repressor on downstream target gene.Different cell phenotypes and states,in vivo environment,and recruitment of coregulatory factors all associate with BACH1 transcription.The research progress of BACH1 is summarized in the article.
