摘要
以树兰(Epidendrum)茎尖为外植体,利用茎尖脱毒法和病毒抑制剂,研究对建兰花叶病毒(Cymbidium masaic virus,CymMV)和齿兰环斑病毒(Odontoglossum ringspot virus,ORSV)的脱毒效果,并建立树兰脱毒苗快速繁殖技术体系。结果表明,茎尖大小在0.3~0.5 mm之间可达到脱毒的效果,或添加2.0 ml·L^(-1)5%氨基寡糖素也可达到有效的脱毒效果;最适合树兰脱毒苗增殖的培养基是改良的1/2MS+6-BA3.0 mg·L^(-1)+NAA0.3 mg·L^(-1),最适壮苗培养基是改良MS+香蕉泥50 g·L^(-1),最适生根培养基是花宝1号+NAA0.8 mg·L^(-1)+香蕉泥50 g·L^(-1)+活性炭1 g·L^(-1)。
The shoot tips of Epidendrum secumdum were cultured as explants to study the detoxification effect of Cymbidium masaic virus(CymMV)and Odontoglossum ringspot virus(ORSV)by using shoot tip detoxification and virus inhibitors.A rapid propagation technology system of virus-free seedlings was established.The results showed that the virusfree effect could be achieved when the shoot tip size was between 0.3-0.5 mm,or with the addition of 2.0 ml·L^(-1)5%amino oligosaccharide.The most suitable medium for multiplication of virus-free seedlings was improved 1/2MS+6-BA 3.0 mg·L^(-1)+NAA 0.3 mg·L^(-1).The medium suitable for sound seedlings was improved MS+banana 50 g·L^(-1).The suitable rooting medium was Hyponex+NAA 0.8 mg·L^(-1)+banana 50 g·L^(-1)+activated carbon 1 g·L^(-1).
