小麦TaPLD-2基因的克隆及功能分析 被引量:1

摘要 为了寻找耐盐新基因,培育耐盐新品种。本研究以23个小麦品种为材料,对TaPLD-2基因进行克隆及序列多态性分析,并对TaPLD-2基因响应盐胁迫表达模式进行了研究。结果表明,TaPLD-2基因的全长编码框为1301 bp,位于2DS染色体上,编码的蛋白定位于细胞质,为亲水蛋白,无跨膜区域,属于可溶性NAD(P)(H)氧化还原酶家族;TaPLD-2基因受盐胁迫诱导上调表达,表达量在盐胁迫24 h为对照组的1.8倍,48 h达到最高,为对照组5倍。分析表明SNP位点与相对根长、相对株高及K+、Na+含量都存在一定程度的相关性。 The study aims to find new salt-tolerant genes and breed new salt-tolerant varieties. 23 wheat varieties were used as materials to clone and sequence polymorphisms of TaPLD-2 gene, and TaPLD-2 gene expression patterns in response to salt stress was studied. The results showed that the full-length coding frame of TaPLD-2 gene was 1301 bp, located on the 2 DS chromosome, and the protein was a hydrophilic protein and located in cytoplasm. The protein had no transmembrane region and belonged to the soluble NAD(P)(H)oxidoreductase family. The TaPLD-2 gene was induced to up-regulated expression by salt stress, and the expression level was 1.8 times that of the control group at 24 h after salt stress, and reached the highest at 48 h,which was 5 times that of the control group. The analysis result showed that the SNP loci had some relevance to relative root length, relative plant height, and K+and Na+contents of salt-treated plants.
