巴旦木壳色素提取工艺优化及成分初步分析

摘要 在单因素的试验基础上,选择乙醇浓度、料液比、提取时间、提取温度、pH值为考察因素,以色素吸光度为指标,采用二次通用旋转组合设计优化巴旦木壳色素提取工艺,建立回归数学模型,对试验结果进行方差分析和频率分析。结果表明,最佳提取工艺为:乙醇浓度46.00%,料液比1∶40,提取时间2.6h,提取温度80.0℃,pH值11.6;通过3次平行试验得到实际吸光度与预测吸光度的相对标准偏差(RSD)为1.45%,表明所建立数学模型可用于巴旦木壳色素提取工艺优化;经过成分特殊显色反应,说明巴旦木壳乙醇提取物中含有黄酮类为主的多种组分。 On the basis of single factor experiment,ethanol concentration,extraction time,extraction temperature and pH are selected as the investigation factor,and pigment absorbance is used as index to optimize the extraction process of pigment from almond shell,and a regression mathematical model is established to analyze the variance and frequency of experimental results.The optimum extraction process is obtained as follows:ethanol concentration 46.00%,material to liquid ratio 1∶40,extraction time 2.6 h,extraction temperature 80.0℃,and pH value 11.6.The relative standard deviation(RSD)of actual absorbance and predicted absorbance is 1.45%through three parallel experiments,which indicates that the established mathematical model could be used to optimize the extraction process of pigment from almond shell.Through the special color reaction of components,through the special color reaction of components,the water/ethanol extract of almond shell contains various componentsm,but the main is flavonoids.
