蒙药材多叶棘豆不同采收期的槲皮素含量动态变化研究

Study on the dynamic change of quercetin content of Mongolian medicine Oxytropis Myriophylla in different harvest periods

目的:检测不同采收期的人工种植的多叶棘豆中有效成分的含量,确定最佳采收期。方法:采用高效液相色谱法测定多叶棘豆质量标准物质槲皮素的含量。色谱柱为syncronis-C_(18)(250 mm×4.6 mm,5μm);流动相为甲醇-0.4%的磷酸溶液(50∶50);流速为1.0 mL/mim;检测波长为370 nm;柱温为25℃。结果:槲皮素在30.156μg~904.684μg范围内具有良好的线性关系(R^(2)=1),含量动态变化区间3.34 mg/g~5.80 mg/g,槲皮素含量随时间变化趋势有一定规律,在6月初到6月中旬采收的含量相对较高,6月中旬至9月初槲皮素含量逐渐下降。结论:6~9月采收的多叶棘豆槲皮素含量高于标准规定的1.00 mg/g,尤其是6月中旬采收的含量达到了5.80 mg/g,质量最佳,符合蒙医药古籍文献中记载的6月中旬夏季和8月中旬秋季为采收最佳时间,为生产加工优质多叶棘豆原材料提供科学依据。

Objective:To determine the dynamic changes of quercetin content of Oxytropis myriophylla(Pall.)DC.at different harvesting time,so as to determine the best collection period.Methods:The content of quercetin was determined by HPLC.The chromatographic column was syncronis-C_(18)(250 mm×4.6 mm,5μm).The mobile phase consisted of methanol-0.4%phosphate solution(50∶50).The flow rate was 1.0 mL/min.The detection wavelength at 370 nm and the column temperature was 25℃.Results:Quercetin had a good linearrelationship in the range of 30.156μg~904.684μg(R^(2)=1),and the dynamic range of quercetin content was between 3.34 mg/g~5.80 mg/g.Quercetin content showed a correlation with the time:the content of quercetin was relatively high from early June to mid-June,and then decreased gradually from mid-June to early September.Conclusion:Quercetin content of Oxytropis myriophylla harvested from June to September was higher than 1.00 mg/g,among which 5.80 mg/g concentration of Oxytropis myriophylla harvested in the middle of June with the best quality.The optimum experimental harvesting period was in line with the ancient Mongolian medicine book recorded.These current results may provide a scientific basis for the production and processing of high-quality Oxytropis myriophylla raw materials.
