基于数据挖掘和网络药理学的脑中风与藏医“隆”功能调节机理研究

摘要 目的:通过数据挖掘技术推测藏药成方治疗脑中风的用药规律,运用网络药理学方法预测对“隆”具有调节作用的高频药物治疗脑中风的作用靶点及通路,分析其作用机制并对主要病因“隆”的内涵进行解析。方法:在《藏药标准》等书中检索、筛选治疗脑中风的藏药成方,根据《晶珠本草》等整理所含的药物,利用R工具arules包对数据进行关联规则分析。将分析得到的高频药物组合中对“隆”具有调节作用的药物,利用文献挖掘、TCMSP等数据库分析治疗脑中风的藏药各药物靶标相互作用关系及相关通路。采用Cytoscape构建治疗脑中风的调“隆”藏药“化合物-靶点-通路-隆”关系网络图。结果:数据挖掘得到沉香、丁香、诃子、木香、肉豆蔻、乳香共6味调“隆”药物,其作用于MAPK3、PTGS2、MAPK14等关键靶点和c GMP-PKG、PPAR等通路,通过调节血管平滑肌、神经元等功能减轻脑中风症状。推测“隆”的功能与血液的运行、血管的功能、神经元的调节、细胞的增殖及凋亡有着密切的关系。结论:本研究探讨了藏族医药治疗脑卒中的作用机制,推测“隆”的功能与血液的运行、血管的功能、神经元的调节等关系密切,为藏医药治疗脑中风提供科学依据。 Objective:To infer the medication rule of Tibetan Medicine in the treatment of cerebral apoplexy by data mining technology,to predict the target and pathway of medicine of regulate rLung with high-frequency in the treatment of cerebral apoplexy by using network pharmacology technology,to analyze its mechanism of action,and to analyze the connotation of the main etiology rlung.Methods:Search and screen the Tibetan prescriptions for stroke in the standard of Tibetan medicine and other books,according to Jingzhu Materia Medica and so on,sort out the medicine contained,using R tool arules package to analyze association rules of data.Among the high-frequency medicine combinations obtained from the analysis,the medicine that have the regulatory effect on rLung are selected,using literature mining,TCMSP and other databases to analyze the interaction and related pathways among the medicine targets of Tibetan medicine in the treatment of stroke.Using the software of Cytoscape to construct the"Compound-Target-Pathway-rLung"network diagram of Tibetan Medicine of regulate rLung for the treatment of stroke.Results:A total of 6 kinds of Tibetan Medicine of regulate rLung including Aquilaria,clove,Terminalia chebula,Aucklandia,nutmeg and frankincense were obtained by data mining,which acted on key targets and pathway such as MAPK3,PTGS2,MAPK14 and cGMP-PKG,PPAR signaling pathway,regulating vascular smooth muscle and neurons to relieve stroke symptoms.It is speculated that the function of rLung is closely related to the operation of blood,the function of blood vessels,the regulation of neurons and the proliferation and apoptosis of cells.Conclusion:This study explored the multi-component,multi-target and multi-channel mechanism of Tibetan medicine in the treatment of stroke,It is speculated that the function of rLung is closely related to the operation of blood,the function of blood vessels and the regulation of neurons,objective to provide scientific basis for the clinical application of Tibetan medicine in the treatment of stroke.
