摘要
目的:使用网络药理学的方法研究蒙药草果四味汤散治疗偏头痛的作用机制。方法:运用TCMSP、GeneCards、DisGeNET和OMIM数据库,分别筛选蒙药草果4味汤散中四味组成药物的活性成分、靶点及疾病靶点,将药物靶点与疾病靶点取交集,得到蒙药草果四味汤散治疗偏头痛的潜在靶点,并构建药物(蒙药草果四味汤散)-活性成分-交集靶点-疾病(偏头痛)相互作用网络图及蛋白相互作用网络图(PPI),并通过网络拓扑分析筛选核心靶点,并对核心靶点在DAVID数据库进行GO功能富集分析、KEGG通路富集分析。结果:筛选得到蒙药草果四味汤散治疗偏头痛的交集靶点117个,经网络拓扑分析,发现蒙药草果四味汤散治疗偏头痛的核心靶点为AKT1,VEGFA、PTGS2等;主要改变的信号通路为AKT信号通路、FoxO信号通路等。结论:蒙药草果四味汤散治疗偏头痛的作用主要通过多个活性成分,作用于多个靶点,影响多条相关通路,发挥消除炎症反应、抑制脑部组织细胞的凋亡、调控血管舒缩功能等作用,为深入探究蒙药草果四味汤散治疗偏头痛的作用机制提供了理论思路和引导方向。
Objective:Using the Mechanism of Mongolian Medicine Caoguo Siwei Tang San in Treating Migraine with the Method of Network Pharmacology.Methods:The TCMSP,GeneCards,DisGeNET and OMIM databases were used to screen the active components,targets and disease targets of the four ingredients of the Mongolian medicine Caoguo Siwei Tang powder,respectively.The drug targets and disease targets were intersected to obtain the potential targets of the Mongolian medicine Caoguo Siwei Tang powder in treating migraine,The interaction network diagram of drug(Mongolian medicine Caoguo Siwei Tang Powder)-active ingredient-intersection target-disease(migraine)and protein interaction network diagram(PPI)were constructed,and core targets were screened through network topology analysis,and GO function enrichment analysis and KEGG pathway enrichment analysis were carried out for core targets in the DAVID database.Results:117 cross targets of Mongolian medicine Caoguo Siwei Tang San for migraine were screened.Through network topology analysis,it was found that the core targets of Mongolian medicine Caoguo Siwei Tang San for migraine were AKT1,VEGFA,PTGS2,etc;The main changed signal paths are AKT signal path,FoxO signal path,etc.Conclusion:The effect of Mongolian medicine Caoguo Siwei Tang San on migraine is mainly through multiple active ingredients,acting on multiple targets,affecting multiple related pathways,playing the role of eliminating inflammatory reaction,inhibiting apoptosis of brain tissue cells,regulating vasomotor function,etc.,which provides theoretical thinking and guiding direction for further exploring the mechanism of Mongolian medicine Caoguo Siwei Tang San on migraine.
