聚丝蛋白基因多态性对EB病毒感染鼻咽癌易感性的影响

Influence of filaggrin gene polymorphism on the susceptibility of nasopharyngeal carcinoma infected by EB virus

摘要 目的探讨聚丝蛋白基因(FLG)多态性对EB病毒(EBV)感染鼻咽癌(NPC)易感性的影响。方法选择湖州市中心医院介入放疗科2018年6月-2020年12月收治的鼻咽癌患者150例作为研究组,选择同期于医院进行体检健康人80名作为对照组。选择FLG基因rs3126085位点作为目标位点设计特异性引物,离心柱法提取全血基因组DNA,采用聚合酶链式扩增反应扩增目的片段,以测序确定分型结果。结果研究组与对照组FLG基因rs3126085位点研究组GG基因型、G等位基因频率为30.67%、49.00%,高于对照组(P<0.05),两组AA基因型、AT基因型频率比较无统计学差异;GG型鼻咽癌患者EBV-DNA阳性人数占比高于AA/AG型(P<0.05),两组临床分期、化疗疗效比较差异无统计学意义;多因素回归分析结果表明,携带FLG基因rs3126085位点GG基因型和G等位基因均是发生鼻咽癌的独立危险因素(P<0.05)。结论FLG基因rs3126085位点GG基因型、G等位基因频率可能增加了本地区居民对鼻咽癌的易感性,其筛查成本效益有待进一步探讨。 OBJECTIVE To explore the effect of filaggrin(FLG)polymorphism on the susceptibility of nasopharyngeal carcinoma(NPC)infected by Epstein-Barr virus(EBV).METHODS A total of 150 nasopharyngeal cancer patients admitted to the Interventional Radiotherapy Department of Huzhou Central Hospital from Jun 2018 to Dec 2020 were enrolled.And 80 healthy people who underwent physical examinations in the hospital during the same period were as the control group.Specific primers for FLG gene rs3126085 site were designed,the whole blood genomic DNA was extracted by spin column method,and the target fragments were amplified by polymerase chain amplification reaction assay.The fragments were confirmed by sequencing.RESULTS The frequency of GG genotype and G allele in FLG gene rs3126085 locus in the study group were 30.67%and 49.00%,respectively,which were higher than those in the control group(P<0.05).There was no significant difference in the frequency of AA genotype and AT genotype between both groups.The proportion of EBV-DNA-positive patients with GG type nasopharyngeal carcinoma was significantly higher than that with AA/AG type(P<0.05).There was no significant difference in clinical staging and chemotherapy efficacy between the two groups.The results of multivariate regression analysis showed that gene rs3126085 locus carring FLG The GG genotype and G allele were independent risk factors for nasopharyngeal carcinoma(P<0.05).CONCLUSION The GG genotype and G allele frequency at the FLG gene rs3126085 may increase the susceptibility of nasopharyngeal carcinoma in local residents,and the cost-effectiveness of screening needs to be further evaluated.
