摘要
目的探讨下调DJ-1基因对甲状腺乳头状癌细胞体外增殖和侵袭能力的影响。方法下调人甲状腺乳头状癌细胞TPC-1中DJ-1的表达,应用Western印迹法检测细胞经过转染前后DJ-1蛋白表达水平;通过迁移和侵袭实验检测下调DJ-1表达前后甲状腺乳头状癌细胞发生迁移和侵袭行为的情况。结果转染si DJ-1组DJ-1蛋白的相对表达量明显较空白对照组和转染非特异性对照组减弱(P<0.05),而后两者之间比较,DJ-1蛋白相对表达量相似,差异无统计学意义(P>0.05)。在细胞迁移和侵袭实验中,转染si DJ-1组中迁移穿膜和侵袭穿膜的细胞数量均明显少于空白对照组和转染非特异性对照组(t迁移=4.494,4.481,t侵袭=4.039,3.594,P<0.05),而空白对照组和转染非特异性对照组之间,差异无统计学意义(t=2.068,2.141,P>0.05)。结论DJ-1基因在人甲状腺乳头状细胞迁移、侵袭中发挥着重要的作用。
Objective To investigate the effect of down-regulation of DJ-1 gene on the proliferation and invasion of thyroid papillary carcinoma cells in vitro.Methods The expression of DJ-1 in human thyroid papillary carcinoma cell line TPC-1 was down regulated,and the expression level of DJ-1 before and after transfection was detected by Western blotting immunoblotting;The effect of down-regulated DJ-1 on the migration and invasion of thyroid papillary carcinoma cells was detected by migration and invasion experiment.Results The relative expression of DJ-1 protein in transfected si DJ-1 group was significantly lower than that in blank control group and transfected nonspecific control group(P<0.05).The relative expression of DJ-1 protein was similar between the latter two groups,and the difference was not statistically significant(P>0.05).In the experiment of cell migration and invasion,the number of migrating and invading cells in the transfected Si DJ-1 group was significantly less than that in the blank control group and the transfected non-specific control group(t migration=4.494,4.481,t invasion=4.039,3.594,P<0.05),but there was no significant difference between the blank control group and the transfected non-specific control group(t=2.068,2.141,P>0.05).Conclusion DJ-1 gene plays an important role in the migration and invasion of human thyroid papillary cells.
