Effect of 7,8-dihydroxyflavone on Proliferation and Apoptosis of Human Cortical Neurons and its Clinical Significance

摘要 目的探讨DHF对ICH神经元的保护效果和分子机制,为缓解ICH损伤,维持神经元功能提供实验依据。方法体外培养的人脑皮层神经元细胞分为对照组,DHF组和Akt抑制剂组,CCK-8、流式细胞凋亡实验和ELSIA检测各组人皮层神经元细胞增殖A值和凋亡细胞数目变化以及Akt、磷酸化Akt(p-Akt)含量。结果相对于对照组,DHF组人脑皮层神经元细胞增殖A值升高,而凋亡数目显著降低(P<0.01);Akt、p-Akt蛋白明显高表达(P<0.01)。与DHF组相比,Akt抑制剂组人脑皮层神经元细胞增殖A值降低,凋亡数目明显升高(P<0.01);Akt、p-Akt蛋白含量显著降低(P<0.01)。结论DHF具有促进人皮层神经元细胞增殖,抑制细胞凋亡的作用,能够发挥保护神经元细胞的良好作用。 Objective To investigate the protective effect and molecular mechanism of DHF on ICH neurons,and to provide experimental basis for alleviating ICH injury and maintaining neuronal function.Methods Human cortical neurons cultured in vitro were divided into control group,DHF group and Akt inhibitor group.CCK-8,flow cytometry and ELSIA were used to detect the proliferation of human cortical neurons,the number of apoptotic cells and the contents of Akt and phosphorylated Akt(p-Akt).Results Compared with the control group,the proliferation a value of cerebral cortical neurons in DHF group increased,while the number of apoptosis decreased significantly(P<0.01);Akt and p-Akt protein were highly expressed(P<0.01).Compared with DHF group,Akt inhibitor group had lower proliferation value and higher apoptosis number(P<0.01).The protein contents of Akt and p-Akt were significantly decreased(P<0.01).Conclusion DHF can promote the proliferation of human cortical neurons and inhibit apoptosis,which can play a good role in protecting neurons.
