黄连解毒汤对APP/PS1小鼠海马记忆功能的影响 被引量:2

摘要 目的通过观察黄连解毒汤(Huanglian Jiedutang,HLJDT)对APP/PS1双转基因小鼠行为学以及白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)表达的影响,探讨HLJDT对阿尔茨海默病(AD)的作用机制。方法以5月龄的APP/PS1双转基因小鼠为研究对象,设为模型组、HLJDT组(433g·kg^(-1))处理,对照组为5月龄同背景小鼠,每组各8只。采用Morris水迷宫和新物体识别实验检测HLJDT对AD小鼠学习记忆能力的改善;采用实时荧光定量聚合酶链式反应(RT-qPCR)检测IL-1β和TNF-αmRNA表达水平。结果HLJDT可以改善APP/PS1双转基因小鼠空间及非空间学习记忆能力,并降低IL-1β和TNF-αmRNA表达水平(P<0.05)。结论HLJDT的抗炎功效,可能是通过调控IL-1β和TNF-α的表达实现的。 Objective To observe the effect of Huanglian Jiedutang(HLJDT)on the behavior of APP/PS1 double transgenic mice and the expression of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α),and to investigate the mechanism of HLJDT in the treatment of Alzheimer’s disease(AD).Methods Five-month-old double transgenic APP/PS1mice were set as model group,HLJDT group(433g·kg^(-1)),and 5 months old mice with the same background were set in the control group,8 cases in each group.Morris water maze and new object recognition experiment were used to test the effect of HLJDT on the learning and memory ability of AD mice.Real-time fluorescent quantitative polymerase chain reaction(RT-qPCR)was used to detect TNF-αmRNA expression levels of IL-1βand TNF-α.Results HLJDT could improve the spatial and non-spatial learning and memory abilities of APP/PS1 double transgenic mice,and reduce the expression levels of IL-1βand TNF-αmRNA(P<0.05).Conclusion The anti-inflammatory effect of HLJDT may be achieved by regulating the expression of IL-1βand TNF-α.
