摘要
目的探讨姜黄素类似物H7对前列腺癌PC3细胞增殖和凋亡的影响。方法采用MTT法检测不同浓度H7在作用不同时间后对PC3细胞增殖情况的影响;分别用姜黄素、AR抑制剂、H7处理PC3细胞24 h,设置空白对照组,采用蛋白印迹法和q-PCR方法检测各组PC3细胞中凋亡蛋白和mRNA表达变化。结果姜黄素类似物H7对PC3细胞增殖有抑制作用;与空白对照组比较,姜黄素组、AR抑制剂组和H7组PC3细胞中Caspase-3、Caspase-9蛋白和mRNA表达显著升高(P<0.05或P<0.01),且H7组升高更明显。结论姜黄素类似物H7可以抑制前列腺癌PC3细胞增殖,并可通过上调PC3细胞中Caspase-3、Caspase-9蛋白和mRNA表达诱导细胞凋亡。
Objective To investigate the effect of curcumin analog H7 on the proliferation and apoptosis of prostate cancer PC3 cells.Methods The effect of different concentrations of H7 on the proliferation of PC3 cells was tested by MTT method.PC3 cells were treated with curcumin,AR inhibitor,and H7 for 24 hours,and a blank control group was set.Western blot and q-PCR were used to detect the changes of Caspase-3,Caspase-9 protein and mRNA expression in PC3 cells of each group.Results Curcumin analogue H7 can inhibit the proliferation of PC3 cells.Compared with the blank control group,the expression of Caspase-3,Caspase-9 protein and mRNA in PC3 cells of curcumin group,AR inhibitor group and H7 group was significantly increased(P<0.05 or P<0.01),and the increase was more pronounced in the H7 group.Conclusion Curcumin analogue H7 can inhibit the proliferation of prostate cancer cell PC3 cells,and can induce apoptosis by up-regulating the expression of Caspase-3,Caspase-9 protein and mRNA in PC3 cells.
