摘要
目的对透骨草(Phryma leptostachya L.)全草乙酸乙酯萃取物的化学成分进行研究。方法采用硅胶柱色谱、ODS柱色谱、凝胶柱色谱、HPLC等色谱手段进行分离纯化,并结合核磁共振波谱技术鉴定化合物的结构,采用MTT法对分离得到的化合物进行H2O2诱导的SH-SY5Y细胞损伤的神经保护活性研究。结果分离并鉴定了8个已知化合物,分别为苜蓿素(1)、salcolin A(2)、salcolin B(3)、异落叶松树脂醇(4)、倍半西班牙冷杉醇B(5)、咖啡酸甲酯(6)、4-[(3,4-二羟基肉桂酰基)-氧基]肉桂酸甲酯(7)、5-(甲氧基甲基)-1-(4-羟基苯乙基)-1H-吡咯-2-甲醛(8)。H2O2诱导的SH-SY5Y细胞损伤的神经保护活性结果显示,在50μmol·L-1浓度条件下,当阳性对照药维生素E的细胞存活率为76.55%时,化合物1和3的细胞存活率为76.47%、77.81%,与维生素E相近;化合物2、6和7的细胞存活率分别为81.68%、92.15%、93.19%,细胞存活率明显增强。结论化合物1~6、8为首次从透骨草属中分离得到,并且化合物1~3、6、7对H2O2诱导的SH-SY5Y细胞损伤具有显著的神经保护活性。
To study the chemical constituents of the ethyl acetate extract of Phryma leptostachya L.,eight compounds were isolated via silica gel column chromatography,ODS column chromatography,gel column chromatography,HPLC and other methods.The structures of these compounds were identified as tricin(1),salcolin A(2),salcolin B(3),isolariciresinol(4),sesquipinsapol B(5),methyl caffeate(6),4-[(3,4-dihydroxycinnamoyl)-oxy]-methyl cinnamate(7),5-(methoxymethyl)-1-(4-hydroxyphenethyl)-1H-pyrrole-2-carbaldehyde(8),respectively.The results of neuroprotective activity of H2O2-induced SH-SY5 Y cell injury showed that when the cell viability of positive control vitamin E was 76.55%(50μmol·L-1),the cell survival rates of compounds 1 and 3 were 76.47%and 77.81%(at the concentration of 50μmol·L-1),respectively.The cell viability of compounds 2,6 and 7 were 81.68%,92.15%and 93.19%(at the concentration of 50μmol·L-1),respectively.As a result,compounds 1-6,8 were isolated from Phryma L.for the first time,and compounds 1-3,6 and 7 showed significant neuroprotective activity of H2O2-induced SH-SY5 Y cell injury.
