摘要
熊蜂是重要的传粉昆虫,在促进农业生产及维护生态系统平衡等发面发挥着重要作用。Smr基因编码的蛋白为SMRTER(SMRT-related and ecdysone receptor interacting factor),是一个转录因子。在模式生物中发现,Smr基因参与调控包括器官发育、代谢及其他多个重要的生物学过程。本研究利用基因合成技术获得熊蜂Smr基因的编码序列,并把它克隆进原核表达载体进行诱导表达,产生SMRTER重组蛋白。然后,利用纯化的SMRTER蛋白制备兔抗SMRTER多克隆抗体。进一步的检测发现,此多克隆抗体对SMRTER蛋白具有良好的特异性。该研究对于更好理解Smr基因在熊蜂中所发挥的生物学功能具有重要的意义。
Bumblebees are important pollinators.Smr gene encodes SMRTER(SMRT-related and ecdysone receptor interacting factor).The gene is involved in many important biological processes,including reproductive organ development,metabolism and neurogenesis.In this study,the coding region of bumblebee Smr gene was obtained by gene synthesis technique,which was then inserted into bacterial expression vector.Next,we induced the expression of Smr gene and its protein products were purifi ed.Then,rabbit anti-SMRTER polyclonal antibody was prepared.The polyclonal antibody has good specificity to SMRTER protein.Our study laid foundation for further investigation of Smr gene in bumblebee.
