白术多糖对环磷酰胺诱导的南海麻黄鸡免疫抑制的调节作用 被引量:4

摘要 为研究白术多糖对免疫抑制南海麻黄鸡的拮抗作用,试验以160只非免疫健康1日龄南海麻黄鸡作为研究对象,在日粮中添加200 mg/kg白术多糖,连续饲喂4周,于4日龄注射环磷酰胺6.7 mg/只,连续注射3 d,7、14、21、28日龄采集免疫器官、静脉采血,计算免疫器官指数,用免疫器官制成组织切片观察其形态学变化,ELISA试剂盒检测IL-2、TNF-α、IL-1β、IFN-γ等细胞因子的变化。结果显示:随着日龄的增加,环磷酰胺多糖组的胸腺指数和脾脏指数逐渐高于环磷酰胺组,多糖组的胸腺指数、脾脏指数在整个试验期都高于对照组,在形态学方面则发现环磷酰胺多糖组髓质内胸腺细胞增多,皮质部胸腺细胞致密,脾小结数量增加,直径更大,脾索变粗,脾脏受到的损伤更少;环磷酰胺多糖组IL-2、IL-1β、TNF-α、IFN-γ在整个试验期都高于环磷酰胺组。结果表明在雏鸡日粮中添加白术多糖不但能提高其免疫力,还能较快恢复因环磷酰胺引起的免疫抑制作用。 In order to study by Atractylodes macrocephala polysaccharide antagonism on immune suppression in Nanhai Yellow chicken caused by cyclophosphamide,one hundred and sixty non-immune healthy one-day-age Nanhai Yellow chickens were selected as subjects.The diet was supplemented with 200 mg/kg polysaccharide of Atractylodes macrocephala for 4 weeks.At the age of 4 days,cyclophosphamide was injected at the dosage of 6.7 mg/chicken for 3 consecutive days.At the age of 7,14,21 and 28 days,the immune organs were collected and blood was collected intravenously.The indexes of immune organs were calculated and the morphological changes were observed by tissue sections of immune organs.The changes of IL-2,TNF-α,IL-1β,IFN-γand other cytokines in surum were detected by ELISA.The results showed that the thymus index and spleen index of cyclophosphamide and polysaccharide group were higher than those of cyclophosphamide group with the increase of age.The thymus index,spleen index in polysaccharide group were higher than those in control group during the whole experiment period.In terms of morphology,it was found that the thymocytes in the medulla increased,thymocytes in the cortex became denser,the number of splenic nodule increased,the diameter was larger,the splenic cord became thicker,and the spleen received less damage in cyclophosphamide and polysaccharide group.The levels of IL-2,IL-1β,TNF-αand IFN-γin cyclophosphamide and polysaccharide group were higher than those in cyclophosphamide group during the whole experiment period.It′s indicated that adding Atractylodes macrocephala polysaccharide to diet could not only improve their immunity,but also quickly recover the immunosuppression caused by cyclophosphamide in chickens.
