鸡lncGHR在成肌细胞中的表达及其对增殖分化的调控 被引量:1

摘要 为探索长链非编码RNA(Long non-coding RNA,lncRNA)对家鸡骨骼肌的生长发育的调控,试验基于先前研究发现的包含GHR基因第8外显子和部分第7、8内含子的lncRNA(lncGHR),通过qPCR定量检测鸡lncGHR表达的组织特异性规律,构建lncGHR的过表达载体pcDNA3.1-lncGHR,并将其转染到QM-7细胞系中验证lncGHR的功能。结果表明,lncGHR在胸肌、腿肌中表达量较高,在其他组织中表达量较低,而在鸡原代成肌细胞分化过程中,lncGHR呈现“先下降后上升”的趋势,而且lncGHR可极显著促进成肌细胞分化并抑制成肌细胞增殖(P<0.01)。研究结果为探讨GHR基因和lncRNA的功能提供参考,为研究成肌细胞增殖分化的表观遗传调控提供重要调控因子。 In order to explore the regulation of long non-coding RNA(lncRNA)on the growth and development of skeletal muscle in domestic chickens,a lncRNA(lncGHR)containing exon 8 and part of introns 7 and 8 of the GHR gene was found from previous study.The tissue-specific pattern of chicken lncGHR expression was quantitatively detected by qPCR,and the overexpression vector pcDNA3.1-lncGHR for lncGHR was constructed and transfected into the QM-7 cell line to verify the function of lncGHR.As the results showed,lncGHR expression was higher in pectoral and leg muscles and lower in other tissues,while during chicken primary myoblast differentiation,lncGHR showed a trend of"firstly decrease and then increase",and lncGHR could significantly promote myoblast differentiation and inhibit myoblast proliferation(P<0.01).The results of this study can provide a reference for investigating the function of GHR genes and lncRNAs and provide important regulators for studying the epigenetic regulation of myoblast proliferation and differentiation.
