无量山乌骨鸡胚胎期脂肪组织发育特征分析 被引量:3

摘要 为研究无量山乌骨鸡胚胎期脂肪组织的发育特征,对胚胎期12日胚龄(E12)到出定、检测单位重量脂肪组织DNA含量、HE染色鉴定脂肪组织的形态学变化并结合Image J软件定量分析细胞面积、RT-qPCR检测FAS、ACCα、SREBP-1和ATGL mRNA表达量。结果表明:从E12~E16显著下降(P<0.05);脂肪细胞面积在D0达到了峰值;FAS、ACCα和SREBP-1 mRNA于其他时期(P<0.05)。综上所述:无量山乌骨鸡在胚胎发育后期通过增加脂肪细胞体积而使脂肪组织重量增加,脂肪组织中4个脂代谢相关基因存在不同的表达模式。 The purpose of this study was to investigate the development characteristics of adipose tissue of Wuliangshan Sooty chicken during embryonic period.Samples were taken at embryonic day E12,E14,E16,E18,E20 and hatching day(D0).The weight of embryo and subcutaneous adipose tissue was determined,the DNA content in adipose tissue was detected,the morphological changes of adipose tissue were identified by HE staining,and the cell area was quantitatively analyzed with Image J software,and the mRNA expression levels of fatty acid synthase(FAS),Acetyl CoA carboxylaseα(ACCα),sterol regulatory element binding protein-1(SREBP-1)and adipose triglyceride lipase(ATGL)were detected by RT-qPCR.The results showed that embryo weight and fat weight increased significantly from E12 to E20(P<0.05),and the relative fat weight reached the peak value at E18.DNA content was significantly decreased from E12 to E16(P<0.05);The cell area reached the peak value at D0.The mRNA expression levels of FAS,ACCαand SREBP-1 were firstly increased,then decreased and then increased,and E20 were significantly decreased(P<0.05).The expression of ATGL in D0 was significantly higher than that in other stages(P<0.05).In conclusion,the weight of adipose tissue by increasing the volume of adipose cells at the later stage of embryonic development,and four lipid metabolisation-related genes have different expression patterns in the adipose tissue of Wuliangshan Sooty chicken.
