环状RNA hsa_circ_0001946和hsa_circ_0125589及环境因素与高血压的关联

Correlation of circular RNA hsa_circ_0001946,hsa_circ_0125589 and environmental factors with hypertension

目的探讨外周血淋巴细胞中环状RNA(circRNAs)及环境因素与高血压患病的关联。方法采用病例对照研究的方法,2017年4月至2018年10月在福建医科大学附属第一医院和协和医院选取高血压患者681例及对照485例进行问卷调查和健康体检;随机选取84例高血压患者和84例年龄、性别、婚姻状况等匹配的对照,运用实时荧光定量聚合酶链反应(PCR)检测circRNAs(hsa_circ_0001946、hsa_circ_0125589)的表达情况;采用多因素Logistic回归模型、叉生分析等分析环境因素和circRNA在高血压患病中的联合及交互作用。结果调整年龄、性别、婚姻状况等因素后,超重/肥胖(OR=1.660,95%CI 1.242~2.220)、腹型肥胖(OR=2.165,95%CI 1.543~3.038)、焦虑(OR=2.146,95%CI1.406~3.278)及高血压家族史(OR=4.255,95%CI 3.177~5.699)是高血压患病的危险因素;外周血淋巴细胞中hsa_circ_0001946高表达也是高血压患病的危险因素(OR=4.130,95%CI 1.851~9.214),而hsa_circ_0125589与高血压患病无相关性。超重/肥胖与hsa_circ_0001946高表达同时存在时患高血压的风险是非超重/肥胖且hsa_circ_0001946低表达的13.121倍(95%CI3.893~44.231)。焦虑与hsa_circ_0001946高表达同时存在时患高血压的风险是非焦虑且hsa_circ_0001946低表达的17.779倍(95%CI 1.875~168.608)。结论高血压是环境因素和遗传因素共同作用的结果,hsa_circ_0001946高表达与超重/肥胖、hsa_circ_0001946高表达与焦虑的联合作用可增加高血压的患病风险,而hsa_circ_0125589与高血压患病无关。

Objective To investigate the main environmental factors of hypertension and the relationship between hypertension and circular RNAs in peripheral blood lymphocytes.Methods This was a case-control study.A total of 681hypertension patients and 485subjects without hypertension in the First Affiliated Hospital of Fujian Medical University and Fujian Medical University Union Hospital between April 2017and October 2018were included.All participations completed the questionnaire investigation,physical examination and laboratory detection.Quantitative real-time polymerase chain reaction(PCR)method was used to measure the expression of circRNAs(hsa_circ_0001946,hsa_circ_0125589)in 84hypertensives and 84controls.Multivariate Logistic regression models and crossover analysis were used to analyze the interaction and association between environmental factors and circular RNAs in hypertension.Results After adjusted by gender,age and marital status,overweight/obesity(OR=1.660,95%CI1.242-2.220),abdominal obesity(OR=2.165,95%CI 1.543-3.038),anxiety(OR=2.146,95%CI1.406-3.278),family history of hypertension(OR=4.255,95%CI3.177-5.699),and high expression of hsa_circ_0001946in peripheral blood leukocytes(OR=4.130,95%CI 1.851-9.214)were risk factors for hypertension,while the expression of hsa_circ_0125589 was not related with hypertension.Crossover analysis showed that the risk of hypertension was 13.121times(95%CI 3.893-44.231)in overweight subjects with high hsa_circ_0001946level compared with normal weight subjects with low hsa_circ_0001946level.Meanwhile,compared to subjects without anxiety and low expression of hsa_circ_0001946,the risk of hypertension was 17.779times(95%CI1.875-168.608)in subjects with anxiety and high expression of hsa_circ_0001946.Conclusion Hypertension is the result of both environmental factors and genetic factors.The interation of hsa_circ_0001946high expression and overweight or anxiety may increase the risk of hypertension,while the expression of hsa_circ_0125589 is not related to hypertension.
