HOOK1在结直肠癌中的表达及其临床意义

Expression of HOOK1 in colorectal carcinoma and its clinical significance

目的探讨HOOK1在结直肠癌中的表达水平及其与患者临床病理特征的关系。方法结合前期生信分析结果,运用GEPIA数据库预测HOOK1在结直肠癌组织和正常结直肠组织中的表达差异及其对预后的影响。收集2021-10—2022-10在扬州大学附属医院行手术治疗的93例结直肠癌患者的癌组织及对应的癌旁组织标本,通过免疫组化方法检测结直肠癌组织及癌旁组织中HOOK1蛋白的表达量,分析其在结直肠癌中的表达水平及其与患者临床病理特征的关系。结果数据库预测结果显示,HOOK1在结直肠癌组织中的表达水平较正常结直肠组织显著升高,且HOOK1高表达组患者的生存率高于低表达组患者(P<0.05)。免疫组化染色结果提示,HOOK1主要表达于细胞质,在结直肠癌组织中的表达显著高于癌旁组织,差异有统计学意义(P<0.001);HOOK1在不同年龄、性别、发生部位、肿瘤大小、TNM分期、脉管侵犯及神经侵犯患者中表达差异无统计学意义(P>0.05);结直肠癌中HOOK1表达与肿瘤浸润深度、淋巴结转移、分化程度密切相关,差异有统计学意义(P<0.05)。结论HOOK1在结直肠癌组织中呈高表达,且HOOK1高表达的患者总体预后较好,提示HOOK1可作为结直肠癌诊疗的一项潜在肿瘤标记物。

Objective To study the expression level of HOOK1 in colorectal cancer and its relationship with clinicopathological features.Methods Combined with the result of early-stage bioinformatics analysis,GEPIA database was used to predict the difference of HOOK1 expression in colorectal cancer tissues and normal colorectal tissues and its influence on prognosis.From October 2021 to October 2022 in the Affiliated Hospital of Yangzhou University,93 paraffin specimens of CRC patients undergoing surgical resection were collected.The expression level of HOOK1 protein in colorectal cancer tissues and adjacent tissues was detected by immunohistochemistry.Its expression level and association with clinicopathological features in colorectal cancer were analyzed.Results The database predicted that the expression level of HOOK1 in colorectal cancer tissues was significantly higher than that in normal colorectal tissues,and the survival rate of patients with high HOOK1 expression was higher than that of patients with low HOOK1 expression(P<0.05).Immunohistochemical staining result showed that HOOK1 was mainly expressed in the cytoplasm,and the expression of HOOK1 in colorectal cancer tissues was significantly higher than that in adjacent tissues(P<0.001).There was no significant difference in HOOK1 expression among patients with different age,sex,tumor location,tumor size,TNM stage of CRC,vascular invasion and nerve invasion(P>0.05).The expression level of HOOK1 was significantly correlated with tumor invasion depth,lymph node metastasis and tumor pathological differentiation(P<0.05).Conclusion HOOK1 is highly expressed in colorectal cancer,and the overall prognosis of patients with high expression of HOOK1 is better,suggesting that HOOK1 can be used as a potential tumor marker in the diagnosis and treatment of colorectal cancer.
