基于DNA条形码技术的仓储皮蠹分子鉴定初步研究 被引量:1

Molecular identification of dermestid beetle adult and larvae in grain depot based on DNA barcoding

摘要 利用DNA条形码技术,选取线粒体DNA细胞色素氧化酶I(COI)基因片段,通过序列测定、比对分析,对采自于我国哈尔滨、长春、酒泉和新乡四个地区粮库和标本馆的皮蠹成虫和幼虫样品进行了分子鉴定。结果表明,在所检测的10头皮蠹幼虫及1头成虫样品中,有5头为黑毛皮蠹(Attagenus unicolor japonicus)、3头为花斑皮蠹(Trogoderma variabile)、2头为百怪皮蠹(Thylodrias contractus)、1头为火腿皮蠹(Dermestes lardarius),与相关种已知序列的相似度在98%以上,其系统发育树明显与我国其他常见皮蠹近缘种区分开。 Dermestid beetles constitute an important class of sto rage pests whose rapid identification is the basis for monitoring and management.In this study,the mitochondrial DNA cytochrome oxidase I(COI)gene fragment was selected by DNA barcode technology.The sequence identification,alignment and analysis were used to identify the adult and larvae samples collected from different areas located in Harbin,Changchun,Jiuquan,and Xinxiang in China.The results showed that among the 10 larvae and 1 adult tested,5 were Artagenus unicolor japonicus,3 were Trogoderma variabile,2 were Thylodrias contractus,1 was Dermestes lardarius,and the similarity of the known sequences was above 98%.These four species were clearly distinguished from other closely related species which often occurred in China shown by NJ phylogenetic tree.
