葡萄籽提取物对小剂量辐射小鼠防护作用的实验研究 被引量:2

EFFECTS OF GRAPE SEED EXTRACT ON LOW DOSE RADIATION-INDUCED DAMAGE IN MICE

摘要 目的观察葡萄籽提取物对小剂量辐射损伤的防护作用。方法24只昆明雄性成年小鼠随机分为空白对照组(NC)、辐射模型组(R)和葡萄籽提取物干预组(G),每组8只。NC组、R组采用蒸馏水连续灌胃21d;G组采用浓度为450mg/(kg bw/d)葡萄籽提取物连续灌胃21d,实验第15d开始,R组、G组接受辐射剂量率为0.08Gy/h/d的X射线照射,每天照1h,连续照射7d,照射总剂量0.56Gy。实验结束后立即处死动物,取脑、肝脏、胸腺、脾脏称重并计算脏器指数;取血检测外周血细胞数量;分离左侧股骨检测骨髓有核细胞数量;试剂盒检测血清、肝组织丙二醛含量和超氧化物歧化酶活性;试剂盒检测血清白细胞介素-2和白细胞白介素-6浓度。结果与NC组比,R组小鼠的外周血白细胞数量、骨髓有核细胞数量、脾指数和胸腺指数显著降低(P<0.05);血清与肝组织丙二醛含量显著升高(P<0.05),超氧化物歧化酶活性显著降低(P<0.05);血清白细胞介素-2浓度显著降低(P<0.05),白细胞介素-6浓度显著升高(P<0.05)。G组与R组比,小鼠的外周血白细胞数量、骨髓有核细胞数量和胸腺指数显著升高(P<0.05);血清、肝组织丙二醛含量降低、超氧化物歧化酶活性升高(P<0.05),血清白细胞介素-6浓度降低(P<0.05)。结论葡萄籽提取物对小剂量辐射损伤具有良好的保护作用,机制可能与增强免疫和抗氧化功能有关. Objective To investigate the effects of grape seed extract on low dose radiation induced damage in mice.Methods Twenty four healthy kunming male adult mice were randomly divided into 3 groups:the blank control group(NC),the radiation model group(R)and the grape seed extract intervention group(G).Mice in G group were gavaged with grape seed extract(450 mg/kgbw)for 21 days.Mice in NC group and R group were treated with distilled water for 21 days,From the 15 th day of the experiment,mice in R and G groups received X-ray irradiation with radiation dose rate of 0.08 Gy/h/d for 1 h every day for 7 d,with a total dose of 0.56 Gy.The animals were sacrificed immediately after the experiment,and the brain,liver,thymus and spleen were weighed and the viscera index was calculated.The number of peripheral blood cells was determined.The left femur was isolated to detect the number of nucleated cells in the bone marrow.Serum and liver malonaldehyde(MDA)content and superoxide dismutase(SOD)activity were detected with assay kits.Serum interleukin(IL-2)and interleukin(IL-6)concentrations were detected with ELISA kits.Results Compared with the NC group,the number of peripheral white blood cells(WBC),bone marrow nucleated cells.spleen index and thymus index were significantly lower in the R group(P<0.05).Serum and liver MDA content were significantly higher(P<0.05)and SOD activity were significantly lower(P<0.05).Serum IL-2 concentration were significantly lower(P<0.05),and IL-6 concentration were significantly higher(P<0.05).Compared with the R group,the number of peripheral WBC,the number of nucleated cells in the bone marrow and the thymus index were significantly higher in the G group(P<0.05);serum and liver MDA content were significantly lower and the SOD activity were significantly higher(P<0.05).Serum IL-6 concentration were significantly lower(P<0.05).Conclusion Consumption of grape seed extract protected mice from low dose radiation induced damage through enhancing immune function and antioxidant function.[ACTA NUTRIMENTA SINICA,2020,42(4):363-368]
