高通量测序研究三种非消化性寡糖及其组合对小鼠肠道菌群的影响 被引量:2

EFFECT OF THREE NON-DIGESTIBLE OLIGOSACCHARIDES AND THEIR COMBINATIONS ON INTESTINAL MICROBIOTA IN MICE EVALUATED BY ILLUMINA HIGH-THROUGHPUT SEQUENCING

摘要 目的研究低聚果糖(Fructooligofructose,FOS)、低聚木糖(xylooligosaccharide,XOS)和低聚半乳糖(galactooligosaccharide,GOS)三种非消化寡糖及其组合对小鼠肠道菌群的影响,方法将140只雌性小鼠随机分为:对照组(空白)、低聚果糖、低聚木糖、低聚半乳糖、低聚木糖与低聚半乳糖(1:1.5)复合、低聚木糖与低聚果糖(2:1)复合、低聚半乳糖与低聚果糖(2:1)复合共7组。各组每天分别以400 mg/kg剂量灌胃,连续15 d。末次灌胃24h后,收集小鼠新鲜粪便,用高通量测序技术分析其中微生物群落组成和结构的变化。结果与对照组相比,试验组小鼠肠道有益菌与致病菌比例差异显著,复合寡糖组小鼠肠道菌群物种多样性和丰富度最高,单一寡糖灌胃组次之;在属水平上,与对照组比,试验组均显著(P<0.05)提高有益菌的物种丰度,其中低聚木糖与低聚半乳糖(1:1.5)复合组、低聚木糖与低聚果糖(2:1)复合组显著(P<0.05)提高乳杆菌属、双歧杆菌属的物种丰度。结论非消化寡糖能够提高小鼠肠道微生物菌群多样性及丰富度,能在一定程度上改善肠道微环境且复合组效果优于单一组。 Objective To study the effects of three non-digestible oligosaccharides-FOS, XOS and GOS-and their combinations on intestinal flora in mice. Methods One hundred and forty female mice were randomly divided into seven groups: control group(blank), XOS, FOS, GOS, XOS and GOS(1:1.5), XOS and FOS(2:1), GOS and FOS(2:1) groups. Results Compared with the control group, the ratios of intestinal beneficial bacteria to pathogenic bacteria in the experimental groups were significantly different. The species diversity and richness of the intestinal flora in the mice of the complex oligosaccharide group were the highest, followed by the single oligosaccharide gavage group. At the genus level, compared with the control group, the abundance of beneficial bacteria was increased significantly in the experimental groups(P<0.05), including XOS and GOS(1:1.5) composite groups as well as XOS and FOS(2:1) composite groups in which the species abundance of Lactobacillus and Bifidobacterium was increased significantly(P<0.05). Conclusion Non-digestible oligosaccharides can improve not only the diversity and richness of intestinal microflora, but also the intestinal microenvironment to a certain extent in mice, and the effect is better in the compound group than in the single group.
