液相色谱串联高分辨质谱Q Exactive Foucs检验血液中常见毒品

Detection of common drugs in blood by Liquid Chromatography Tandem High Resolution Mass Spectrometry Q Executive Foucs

摘要 目的建立一种快速检验血液中10种常见毒品的液相色谱串联高分辨质谱(Q Exactive Foucs)检测方法,研究上述10种常见毒品在液相色谱串联高分辨质谱(Q Exactive Foucs)PRM扫描模式下的定性参数(保留时间、离子碎片)。方法血液样本经过前处理,应用超高液相色谱分离法分离,采用高分辨质谱的平行反应监测模式(PRM)分析,并考察方法的有效性、检出限、回收率和精密度。结果通过对血样中10种不同浓度毒品的添加实验,对方法进行了验证,10种毒品各成分分离良好,检出限在0.25~10 ng/ml;回收率为85.3%~126.8%;重现性和稳定性良好,RSD均小于5%。结论本文方法简单、灵敏、准确、实用,可完成多种毒品筛选、确认只需一次前处理、一个实验条件,大大节省了检测时间和工作量。 Objective To establish a rapid method for the detection of 10 common drugs in blood by liquid chromatography tandem high resolution mass spectrometry(Q Executive Foucs),and to study the qualitative parameters(retention time,ionic fragments)of the above 10 common drugs under the PRM scanning mode of Q Executive Foucs.Methods The blood was pretreated,separated by ultrahigh liquid chromatography,and analyzed by parallel reaction monitoring mode(PRM)of high resolution mass spectrometry.The validity,detection limit,recovery and precision of the method were investigated.Results The method was validated by adding 10 kinds of drugs with different concentrations in blood samples.The components of 10 kinds of drugs were well separated,and the detection limit was 0.25~10 ng/ml;The recovery rate is 85.3%~126.8%;The reproducibility and stability are good,RSD is less than 5%.Conclusion This method is simple,sensitive,accurate and practical.It only needs one pretreatment and one experimental condition to complete the screening and confirmation of multiple drugs,which greatly saves the detection time and workload.
