透骨消痛胶囊抑制大鼠骨性关节炎作用研究

Study on mechanism of Tou Gu Xiao Tong Capsule inhibiting synovitis of osteoarthritis in rats

目的探究透骨消痛胶囊对大鼠骨性关节炎的抑制作用。方法将大鼠随机分成5组:对照组、模型组及透骨消痛胶囊低、中、高剂量(0.93、1.86、3.72 g/kg)组,除对照组外,采用手术方法构建SD大鼠骨性关节炎模型,对照和模型组ig给予等量生理盐水,每天1次,持续8周。给药结束后,大鼠眼眶取血后分离血清,处死大鼠取膝关节软骨。软骨组织HE染色后用改良Mankin评分细则进行评分;实时荧光定量PCR(q RT-PCR)和Western blotting检测软骨中基质金属蛋白酶(MMP)-2和MMP-13的表达;ELASA法检测血清炎性介质的表达。结果与模型组比较,透骨消痛胶囊0.93、1.86、3.72 g/kg剂量组HE染色评分均显著升高(P<0.05),MMP-2、MMP-13 m RNA和蛋白水平均显著降低(P<0.01);血清肿瘤坏死因子-α(TNF-α)、血管内皮生长因子(VEGF)、白细胞介素-6(IL-6)和白细胞介素-1β(IL-1β)水平均显著降低(P<0.01)。结论透骨消痛胶囊可以显著抑制大鼠骨性关节炎,机制与降低MMP-2、MMP-13表达,降低血清炎症因子TNF-α、VEGF、IL-6和IL-1-β水平相关。

Objective To explore the inhibitory Mechanism of Tou Gu Xiao Tong capsule on rat osteoarthritis synovitis.Methods The rats were randomly divided into five groups:control group,model group,low,medium and high dose(0.93,1.86,3.72 g/kg)group of Tou Gu Xiao Tong Capsule.Except the control group,SD rats were operated to construct osteoarthritis model.Rats of control group and model group were ig given the same amount of normal saline once a day for 8 weeks.At the end of administration,blood was collected from the orbit of the rats and serum was separated.After HE staining,the cartilage tissue was graded by modified Mankin scoring rules;the expression of matrix metalloproteinase(MMP)-2 and MMP-13 in cartilage was detected by real-time fluorescent quantitative PCR(q RT-PCR)and Western blotting;the expression of serum inflammatory mediators was detected by elasa method.Results Compared with the model group,the HE staining scores of 0.93,1.86 and 3.72 g/kg groups of tgxtc were significantly higher(P<0.05),the levels of MMP-2,MMP-13 m RNA and protein were significantly lower(P<0.01),the levels of TNF-α,VEGF,IL-6 and IL-1βwere significantly lower(P<0.01).Conclusion Tou Gu Xiao Tong Capsule can significantly inhibit osteoarthritis in rats,the mechanism is related to the decrease of MMP-2,MMP-13 expression,TNF-α,VEGF,IL-6 and IL-1-βlevels.
