茵陈蒿汤物质基准特征图谱的建立

Establishment of Materialenchmark Characteristic Atlas of Yinchenhao Decoction

目的:建立能够反映茵陈蒿汤功效的特征图谱。方法:针对组方中各药味的功效与药理作用相关,筛选特征图谱目标物质,采用HPLC法围绕目标成分建立其特征图谱。色谱条件为:Agilent-C_(18)(250×4.6mm,5μm)色谱柱;流动相:乙腈(A)-0.05%(B)磷酸溶液梯度洗脱;检测波长:254nm;体积流量:1mL·min^(-1 );柱温:30℃;建立共有模式,并进行峰归属及指认,根据反映主要功效的特征峰进行质量评价。结果:建立了茵陈蒿汤物质基准的特征图谱,其中归属茵陈的峰12个、归属栀子的峰2个、归属大黄的峰7个,指认了8个与功效相关的色谱峰,分别为没食子酸、绿原酸、秦皮乙素、栀子苷、金丝桃苷、槲皮素、大黄酸、大黄素。同时关联茵陈蒿汤主要功效,选取17个特征峰,并选择以绿原酸峰为S峰,计算其余特征峰相对保留时间的方式评价茵陈蒿汤物质基准质量。结论:该方法采用特征图谱反映特征成分信息的特点,对功效相关的重点成分进行质控,建立了茵陈蒿汤物质基准的特征图谱,为后续关于茵陈蒿汤制剂的开发及质量控制提供理论依据。

Objective:To establish a characteristic atlas that can reflect the efficacy of Yinchenhao Decoction.Methods:Aiming at the correlation between the efficacy and pharmacological action of each medicine in prescription,the target substances of characteristic map were screened,HPLC was used to establish the characteristic chromatogram around the target components.Chromatographic conditions:Agilent-C_(18)(250×4.6 mm,5μm)column;Mobile phase:gradient elution of acetonitrile(A)-0.05%(B)phosphoric acid solution;Detection wave:254 nm;Volume flow:1 ml/min;Column temperature:30℃;The common mode was established,and the peaks were assigned and identified.The quality evaluation was carried out according to the characteristic peaks reflecting the main efficacy.Results:The characteristic Atlas of material standard of Yinchenhao Decoction was established,Among them,Among them,12 peaks belong to Artemisiae scopariae herba,2 peaks belong to Gardeniae fructus,7 peaks belong to Rhei radix et rhizoma,eight peaks related to efficacy were identified,which were gallic acid,chlorogenic acid,aesculetin,gardenoside,hyperin,quercetin,rhein and emodin.At the same time,according to the main efficacy of Yinchenhao Decoction,17 characteristic peaks were selected,and chlorogenic acid peak was selected as S peak to evaluate the quality of Yinchenhao Decoction Material benchmark by calculating the relative retention time of other characteristic peaks.Conclusion:In this method,characteristic maps reflect the characteristics of characteristic component information,quality control is carried out on key components related to efficacy,and the characteristic maps of Yinchenhao Decoction Material benchmark are established,which provides theoretical basis for the subsequent development and quality control of Yinchenhao Decoction.
