肉桂抗病毒有效部位总糖测定及单糖组成分析 被引量:1

摘要 目的:建立肉桂提取物中总糖的含量测定及单糖组成分析方法。方法:采用硫酸-苯酚比色法,优化实验参数建立分析方法,对3批肉桂提取物进行测定。采用PMP柱前衍生化HPLC法进行单糖组成分析。结果:硫酸-苯酚比色法最优参数为:检测波长为490nm,5%苯酚溶液1mL,浓硫酸5mL。葡萄糖在0.0050~0.0175mg/mL浓度范围内与吸光度呈现良好的线性关系,R^(2)=0.9997。3批肉桂提取物中总糖含量分别为36.12%、39.23%、41.66%。单糖组成分析显示肉桂提取物水解后主要的单糖组成为甘露糖(Man)、鼠李糖(Rha)、葡萄糖(Glc)、半乳糖(Gal)和阿拉伯糖(Ara),摩尔比例分别为0.14∶0.28∶0.38∶1∶0.92。结论:该研究方法准确、灵敏、可靠,可用于肉桂提取物中总糖的含量测定和单糖组成分析。 Objective:To establish a method for determination of total sugar content and analysis of monosaccharide composition in Cinnamomum Cassia extracts.Methods:The sulfuric acid-phenol method was employed,analytical parameters were optimized and the method was applied to the analysis of 3 batches of samples.PMP pre-column derivatization HPLC method was used to determine the monosaccharide composition of Cinnamomum Cassia extracts.Results:The optimal parameters for sulfuric acid-phenol method were as follows:the detection wavelength of 490 nm,5%phenol solution 1 mL,sulfuric acid 5 mL.The linearity range was 0.0050~0.0175 mg/mL,showing good linearity(R^(2)=0.9997).The contents of total sugars in 3 batches of Cinnamomum Cassia extracts were 36.12%,39.23%and 41.66%.The main monosaccharides in cinnamon extracts were mannose,rhamnose,glucose,galactose and arabinose,the molar ratios were 0.14∶0.28∶0.38∶1∶0.92 respectively.Conclusion:The study method is accurate,sensitive,reliable,and could be used in the determination of total sugars and analysis of monosaccharide in Cinnamomum Cassia extract.
