摘要
目的:采用正交设计优化金毛狗脊指纹图谱的前处理方法。方法:在建立同时检测5-HMF、原儿茶酸、原儿茶醛三种成分的基础上,采用正交设计及方差分析,分别研究溶剂类型、溶剂比例、超声时间和次数对上述三个成分得率的影响。结果:最佳处理方法为:甲醇-1%冰醋酸溶液(70∶30)超声处理30min,1次。结论:在最佳工艺条件下,上述三个成分得率为0.381%,高于其他条件下的得率,该处理方法可为狗脊指纹图谱研究提供实验依据。
Objective:Orthogonal design was used to optimize the pretreatment method of the fingerprint of Cibotii rhizoma.Methods:On the basis of simultaneous determination of 5-HMF,protocatechuic acid and protocatechuic aldehyde,the effects of solvent type,solvent ratio,ultrasonic time and frequency on the yield of the three components were studied by orthogonal design and analysis of variance.Results:The optimal conditions of extraction were as follows:to add the methanol-1%glacial acetic acid solution(70∶30)and ultrasonic treatment for 30 min for each time.Conclusion:Under the optimum conditions,the yield of the above three components was 0.381%,which was higher than that under other conditions.This method can provide experimental basis for the fingerprint study of Cibotii rhizoma.
