五加参归芪精改善小鼠心气虚量效关系研究 被引量:2

摘要 目的:研究五加参归芪精改善小鼠心气虚的药效作用及量效关系。方法:采用"基础进食+负重游泳+灌服心得安"方法建立小鼠心气虚模型,设置正常组、模型组、阳性药组及五加参归芪精0.76、1.52、3.03、6.06mL/kg 4个给药剂量组,通过检测小鼠游泳力竭时间、血清白介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)、心肌匀浆中肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、丙二醛(MDA)及心肌病理切片等,考察五加参归芪精改善心气虚的药效作用及量效关系。结果:与模型组相比,各给药剂量组心肌结构损伤均减轻;0.76mL/kg剂量组可显著升高SOD、降低MDA,差异均有统计学意义(P<0.01);1.52mL/kg剂量组可显著降低IL-2、MDA,差异均有统计学意义(P<0.01);3.03、6.06mL/kg剂量组对考察的各项指标均有显著改善作用,可延长游泳力竭时间,降低IL-2、TNF-α、CK、LDH、MDA,升高SOD,差异均有统计学意义(P<0.05或P<0.01)。结论:五加参归芪精具有改善小鼠心气虚的药效作用,在0.76~6.06mL/kg剂量范围内具有量效相关性,在3.03、6.06mL/kg剂量下药效作用显著。 Objective:This work aims to study the dose-effect relationship of Wujiashenguiqijing on improving heart-qi deficiency in mice.Methods:The method of"basic eating+weight-bearing swimming+infusion of propranolol"was used to establish the heartqi deficiency mice model.The experiment set up normal group,model group,positive drug group,and four dose groups(0.76,1.52,3.03,and 6.06 mL/kg)of Wujiashenguiqijing.The efficacy and dose-effect relationship of Wujiashenguiqijing in improving heart-qi deficiency were analyzed by detecting the swimming exhaustion time,serum levels of interleukin-2(IL-2)and tumor necrosis factor-α(TNF-α),myocardial homogenate levels of creatine kinase(CK),lactate dehydrogenase(LDH),superoxide dismutase(SOD),malondialdehyde(MDA),and histopathological status of myocardium.Results:Compared with the model group,the damage of myocardial structure in four dose groups were all alleviated;0.76 mL/kg dose group can significantly increase SOD and decrease MDA,the difference is statistically significant(P<0.01);1.52 mL/kg dose group can significantly reduce IL-2 and MDA,and the difference is statistically significant(P<0.01);3.03 and 6.06 mL/kg dose groups have the significant improvement effect on the all indicators,including prolong the swimming exhaustion time,reduce IL-2,TNF-α,CK,LDH,MDA,and increase SOD.The differences are statistically significant(P<0.05 or P<0.01).Conclusion:Wujiashenguiqijing has the effect on improving heart-qi deficiency in mice.It has a dose-effect correlation in the dose of 0.76~6.06 mL/kg,and the effect is significant at the doses of 3.03 and 6.06 mL/kg.
